diseases as a kind of psychosomatic disease,its incidence has also been rising,the occurrence of a variety of gastrointestinal diseases has seriously affected people’s quality of life.Among them,stress ulcer(SU)caused by the body under long-term or severe stress is one of the most common gastrointestinal diseases.In severe cases,it can lead to large-area ulcers and bleeding.The rate can be as high as 50%or more.Stress ulcer(SU)refers to acute gastritis that occurs during shock,trauma,post-surgery and severe systemic infections.It is often accompanied by hemorrhagic symptoms.At the same time,extensive gastric mucosal congestion,edema,and multiple erosions may occur.Acute gastric mucosal lesions can easily lead to death if the treatment is not timely or is not fully understood.It is a more serious form of acute gastric mucosal lesions.Clinically,it is also known as acute stress mucosal lesion(ASML).At the same time,there is also evidence in modern medical research that the occurrence and development of gastrointestinal diseases are often accompanied by the development of sleep disorders,which is closely related to the deregulation of cerebral cortex function and the strong secretion of epiglottic corticosteroids.In the name of TCM,there is no such thing as " stomach ulcer " but generally it can be classified into "stomachache","noisy"stomach and other diseases in accordance with the clinical symptoms and characteristics.The understanding of whether the spleen and stomach function is normal or not has important effects on sleep,such as the opinion of"The Yellow Emperor’s Internal Classic" that "disharmonious stomach is uncomfortable"and explains why in the "Inversion Theory":"Yangming Gastrointestinal veins,stomach,stomach,six seas,its gas is also down,Yangming inverse must not be from its way,it can not be lying also."under the Jing" said:stomach is not lying and restless,this also said that".Because the acupuncture treatment itself has the functions of dredging the meridians,harmonizing yin and yang,and relieving the evil,it plays an important role in the regulation of autoimmune and endocrine systems.Therefore,this article seeks to examine the effects of acupuncture on stress ulcers and sleep disorders.Improve the situation and strive to provide new ideas,methods and means for clinical treatment.Objective:From the experimental point of view,this study replicated a rat model of stress ulcer by restraint-immersion stress and evaluated the degree of gastric mucosal damage in rats by using the ulcer index(UI).The acupuncture group was established after modeling.Acupuncture at Zhongcuo and Zusanli points was used to intervene.The drug group was treated with omeprazole.The effects of acupuncture on serum and brain CRH and ACTH in rats with gastric ulcer were studied.The relationship between stress ulcer and sleep was further discussed.Relevance and acupuncture treatment for its mechanism and effect.Method:(1)28 male Sprague-Dawley rats were modeled,blank group,acupuncture group and drug group using digital table method,7 mice in each group.(2)The blank group was not modeled,and the remaining three groups were all subjected to restraint-immersion(RWIS)modeling to create a stress ulcer(SU)rat model.On the 3rd day after the first model establishment,blanks were removed.The groups outside the group repeated the above-mentioned modeling process and were again modeled.Intervention was performed on the first day after the model was established.The acupuncture group had acupuncture at Zhongwan and Zusanli points for five consecutive days.The drug group was given 2 ml of omeprazole enteric-coated tablets daily for five consecutive days.(3)Using behavioral indicators to observe the rat’s autonomic activity and the sleep time induced by pentobarbital sodium.Then we investigated the effect of acupuncture on the rat’s autonomic activity and the sleep time induced by pentobarbital sodium.(4)Gastric mucosal injury index was calculated by Guth scoring method to observe gastric pathological morphology under light microscope.Learn to change.(5)The expression level of serum CRH and ACTH,CRH in the hypothalamus,and ACTH in the pituitary were detected by ELISA(enzyme-linked immunosorbent assay).Results:(1)Gastric mucosal ulcer index score:Compared with the blank group,the gastric mucosal ulcer index of the model group was significantly higher(P<0.01);compared with the model group,the gastric mucosal ulcer index was significantly lower in the acupuncture group and the drug group(P<0.05);There was no significant difference between the acupuncture treatment group and the drug treatment group(P>0.05).(2)Sleep duration:Compared with the normal group,the sleep time of the model group rats was significantly shorter(P<0.05);Compared with the model group,the sleep time of the rats in the acupuncture treatment group was significantly prolonged(P<0.01);There was no significant difference between the treatment group and the model group rats;the duration of sleep was significantly increased in the acupuncture treatment group and the drug treatment group(P<0.05).(3)Morphology of the gastric mucosa under the naked eye:In the normal group,the gastric mucosa was smooth and intact and no damage was observed.The rats in the model group had marked gastric mucosal hyperemia.There were a large number of point-like hnear bleeding points and erosions.The lesions were significantly high.In the normal group,the degree of gastric mucosal congestion in the acupuncture treatment group and the drug treatment group was lower than that in the model group,with less punctual linear bleeding points and erosion.(4)Morphology of gastric mucosa under light microscope:compared with the normal group,the gastric mucosa cells in the model group were disorderly arranged,the mucosal epithelium was defective,the gland was disordered or destroyed,the mucosa was congested and edema,and a large number of red blood cells were seen.Out,gastric mucosal cell swelling,nuclear necrosis;compared with the normal group,acupuncture treatment group:most of the gastric mucosa structure is normal,the epithelium is complete,red blood cells oozing out,occasionally a small amount of cell necrosis;compared with the normal group Drug treatment group:The structure of gastric mucosa was basically complete,cells were slightly edema,a few epithelial cells were detached and necrosis,and red blood cells were less effusion.(5)The molecular biological mechanism exploration:① The expression of CRH in the hypothalamus:Compared with the blank group,the model group was significantly elevated(P<0.01);compared with the model group,the content of the acupuncture group was significantly lower(P<0.01);the content of the drug group was decreased.The difference was statistically significant(P<0.05);there was no significant difference between the acupuncture group and the drug group(P>0.05).② The expression of serum CRH:compared with the blank group,the model group was significantly higher(P<0.01);compared with the model group,the content of the acupuncture group was significantly lower,the difference was statistically significant(P<0.01);the drug group content was reduced,but No statistical difference(P>0.05);acupuncture group compared with the drug group,acupuncture group content was lower,the difference was statistically significant(P<0.05).③ The expression of ACTH in the pituitary:Compared with the blank group,the model group significantly increased(P<0.01);compared with the model group,the acupuncture group,the drug group content decreased,although there was no statistical difference(P>0.05),but there is a downward trend There was no significant difference between the acupuncture group and the drug group(P>0.05).④The expression of serum ACTH:Compared with the blank group,the model group was significantly increased(P<0.01);Compared with the model group,the acupuncture group content was significantly reduced,the difference was statistically significant(P<0.01),the drug group content was reduced,but No statistical difference(P>0.05);acupuncture group compared with the drug group,acupuncture group content was lower,the difference was statistically significant(P<0.05).Conclusion:1.gastric ulcer can affect sleep in rats,and acupuncture has a certain effect on improving sleep quality.2.acupuncture "Zhongli","Zusanli" can reduce the area of ulcers,improve ulcer inhibition rate,protect the gastric mucosa.3.After restraint-immersion stress modeling,the rat’s sleep time induced by pentobarbital sodium can be significantly shortened.4.The formation of stress gastric ulcer is related to the increase of CRH and ACTH levels.Acupuncture can effectively intervene and the effect is better than that of omeprazole.5.One of the mechanisms of acupuncture acupuncture to increase the inhibitory rate and durationof
