Acupuncture Experimental Study Of The Effects Of The Ethanol Model Of Gastric Ulcer Rat Tnf-¦Á

Gastric ulcer (GU), as one kind of peptic ulcer (PU), is a clinical common, frequently-occurring and chronic disease. The most common clinical symptoms are epigastric pain, bloating, eructation, pantothenic acid, loss of appetite and indigestion. GU is caused by over secretion of acid and pepsin or weak immunity of gastric mucosa or both of the two mentioned reasons.When GU deteriorates, complications such as bleeding, perforation, pyloric obstruction, and cancer may occur.The correlation of stomach and sleep has been discussed in says:"Discomfort of stomach causes disturbed sleep." According to traditional Chinese medicine scholars, stomach digests food, spleen circulates the digested food and then makes it into Qi and blood. Thus, stomach and spleen is material base of human body. Over-eating, unhappiness and long-lasting fatigue can cause dysfunctions of stomach and spleen. Further affect the circulation of Qi which can cause sleeping problem.Acupuncture can dredge the meridian and reconcile yin-yang. Recent studies show that acupuncture can play a two-way adjusting role through multi-level and various ways and methods in improving heath status. It can improve the immune system by affecting the release of cytokines. Studies also show that acupuncture has good effect on the multiplication of epithelial cells and the recovery of gastric mucosa and gastric wall. Meanwhile, can improve sleep quality and increase sleep time.Objective:In this experiment, we feed rats with ethanol to cause gastric ulcer and put needle on "zhongwan", " zusanli", " shenmai" and" zhaohai". We choose capabil ity of action changes of each group, changes of the form of gastric mucosa, influence on sleep time and changes of serum TNF-αcontent as indicators. The objective of this study is to explore the mechanism of using acupuncture to cure GU and the correlation of GU and sleep.Methods:We chose 40 healthy male Wistar rats and after they adapted to the environment, divided them into normal group, GU group, treatmentⅠⅡ,Ⅲgroup and 8 rats for each group. We used ethanol to create GU rats and then treated the cure groups with acupuncture for 5 days. After that, we did open field test and sleep time test first and after finishing the two tests, we killed the rats to measure the changes of form of gastric mucosa and the changes of serum TNF-a.Results: 1. Capability of action:In open field test, GU group walked farther than treatment groups and the normal group with highly significant differences (P< 0.01). Normal group showed significant differences with treatmentⅠ,Ⅱgroup (P<0.05). No difference found between normal group and treatment III group.2. Gastric mucosa damage indicator:GU group and treatmentⅠ,Ⅲgroups showed significant differences (P<0.01). GU group and treatment II group also showed significant difference (P<0.05).2) Sleep time (Pelltobarbitalum Natricum):GU group showed highly significant differences with normal group and the treatment III group (P<0.01) and was significantly different fromⅠ,Ⅱgroup (P<0.05).4. TNF alpha content:Normal group was significant different with GU group (P<0.05) and GU group also showed significant difference with treatment groups (P<0.05). Especially with group III, difference was significant at P<0.01 level. GroupⅠandⅢalso showed significant difference (P<0.05).Conclusion:1. Acupuncture can effectively improve the mental state of AU rat and its capability of action.2. Acupuncture can relive ulcer inflammatory exudate and bleeding. Also it can help gastric mucosa repair.3. Acupuncture can prolong the sleep time and improve sleep quality.4. Acupuncture can reduce serum TNF alpha content, which means acupuncture can promote gastric ulcer cure by adjusting immune system.In summary, this experiment proved acupuncture has a curative effect on gastric ulcer and the correlation between stomach and sleep, which provides experimental basis for further research on mechanism of "uncomfortable sleep causes disturbed sleep...
