| Androgenetic alopecia(AGA)is a common clinical alopecia disease.In China,the epidemiology of this disease is:male prevalence rate is 21.3%,female prevalence rate is 6.0%.AGA often occurs during or after puberty,showing a gradual decrease in hair.Male mainly has hair loss from two temples and gradually extend to the entire scalp,or from the top of the head and gradually extend to the forehead.Female’s symptoms usually performance as hair thinning and gradually extend to temporal and occipital,but women are generally not accompanied by hairline retraction.Androgenetic alopecia causes changes in the appearance of the patient.As the disease gradually develops with time,the amount of hair gradually decreases until it is completely bald.Bringing great mental stress to patients,causing troubles for patients’lives and work.Therefore,it is necessary to creat a new topical drug with small side effects,using convenient and cheap.This article is mainly divided into two parts:The first part is a literature review.Literature Review 1 is the chemical composition、pharmacological action and clinical application of traditional Chinese medicine for the treatment of androgenetic alopecia.The latest research progress of several traditional Chinese medicines summarized in the literature is summarized.And explore the link between its pharmacological effects and treatment of hair loss.Literature review 2 is the research progress of C57BL/6 mouse hair loss model.By summarizing the literature,the application of C57BL/6 mice to study the process steps of hair loss diseases,animal and drug selection,observation indicators were summarized.The second part is the clinical trial and animal experiment.Objective:1.Clinical trial:To evaluate the efficacy and safety of a kind of Chinese herbal gel in the treatment of AGA by using hair analytical system.2.Animal experiment:Using C57BL/6 mice as the research target,inducing its hair into growth phase,explores the effect of the Chinese herbal gel on the hair growth of mice.Method:1.Clinical trial:Randomized single-blind and autologous control methods were used.69 patients with androgenetic alopecia were randomly divided into Chinese herbal gel treatment group and blank gel treatment group.The patients were randomly assigned to Chinese herbal gel group of 46 people in a ratio of 2:1,and the blank gel group covers 23people.All the male patients were asked to smear their own gels on the scalp of both side of the frontal angle every day(female patient smear the gel in the middle of the top of the head).The patient was reviewed e’very 4 weeks.They need to fill out the case report form,get the hair microscope examination,and take clinical photograph.The observation time is 6 months.At the end of the trail,the results of hair microscope could be used to study the hair rate,total number of hairs,scalp inflammation,and patient’s subjective feelings(according to the contents in the case report form),all those results were used to evaluate the clinical and the efficacy of traditional Chinese herbal gel treatment of AGA.2.Animal experiment:24 mice were randomly divided into 3 groups.The three groups were:minoxidil group,normal saline group,and Chinese herbal gel group.The hair in the 2 cm×4cm area of the back of the mouse were removed by using depilatory cream with the purpose of inducing hair follicles in the hair removal zone into the growth phase.On the next day,smear normal saline,5%minoxidil and Chinese herbal gel on the depilatory area for each group.Changes in skin color and hairiness of the mice were observed daily as well as the changes of skin color and the length of hair growth.On the 9th day,3mice in each group were killed and get the skin of the depilatory area to make HE staining.On the 18th day,the remaining 5 mice in each group were also killed and making HE staining.Observe the thickness of the dermis.Ten hairs were taken from the same part of each mouse and the length was measured.Result:1.Clinical trial:Compared with before and after treatment,the hair variation rate in the experimental group decreased by 3%on average after treatment(P<0.05).In scalp inflammation,after treatment,the average reduction was 0.56(P<0.05).In scalp pruritus degree,the average improvement was 0.33 points after treatment(P<0.05).In terms of scalp oil level,the average improvement was 0.31 points after treatment(P<0.05).In folliculitis,the average improvement was 0.53 points after treatment(P<0.05).There were no significant differences in hair variation rate,scalp inflammation,oil improvement,pruritus and folliculitis improvement before and after treatment in the control group(P>0.05).The hair growth rate(P>0.05),scalp inflammation(P<0.05),lipid improvement(P<0.05)and satisfaction(P<0.05)of the experimental group were better than those of the control group.There was no significant difference in hair variation rate,itching degree and folliculitis between the two groups(P>0.05).In terms of safety,there were no adverse reactions in the experimental group and the control group.2.Animal experiment:There were no changes in the skin of each group,such as damage,edema,redness,and exudate.According to the statistical results,there was no significant difference in the thickness of dermis between the Chinese herbal gel group and the minoxidil group on the 9th day(P>0.05),which was statistically different from the saline group(P<0.05).On the 9th day,the growth of hair follicles in the Chinese herbal gel group and the minoxidil group was better than that in the saline group,and the situation was similar between the two groups.There was no statistical difference in dermal thickness between the three groups on the 18th day.There was no statistical difference in the number of hair follicles between the two time points.In terms of hair length,there was no significant difference between the minoxidil group and the Chinese herbal gel group(P>0.05).The two groups were statistically significant compared with the saline group(P<0.05).The Chinese herbal gel group promotes hair growth and is similar to minoxidil.The Chinese herbal gel is safe.Conclusion:1.Clinical trial:Compared with before and after treatment,the experimental group showed that the hair growth rate,scalp inflammation,itching,lipid level and folliculitis improved after treatment were better than those before treatment.After comparing the two groups,the test group was better than the control group in terms of hair growth rate,scalp inflammation,lipid level improvement and satisfaction.The Chinese herbal gel has good safety.2.Animal experiment:Chinese herbal gel has a certain hair growth effect,similar to the effect of minoxidil,and has good safety.Significance:Chinese herbal gel has a good effect in the treatment of androgenetic alopecia,and no adverse reactions.It is worthy of promotion and application in the clinic. |
