Study On The Quality Standard And Pharmacokinetics Of The Alcohol Extract Of Radix Paeoniae Alba

Purpose:In this study,Zhang Zhongjing's"Treatise on"recorded in the GaN based on herbs of Radix Paeoniae Alba,application of modern science and technology and means of alcohol extract of Radix Paeoniae Alba Radix Glycyrrhizae,identification and comprehensive evaluation on the TLC and fingerprint method,through to the peony ester glycosides,paeoniflorin,liquiritin,ISO glycyrrhizic acid,liquiritin and isoliquiritigenin as index components for content determination,the establishment of quality standards,thus a comprehensive and effective evaluation of its quality and to provide reference for the improvement of the quality evaluation system;based on herbal extracts of Paeonia and glycyrrhizina alcohol serum fingerprint and pharmacokinetic study,investigation into the the blood components,and to study the related indexes and pharmacokinetic parameters,the metabolic processes in vivo,to explore the material basis of its efficacy.Thus,the best polar extract part of paeony,licorice and ethanol is elucidated scientifically and scientifically,which provides a more scientific and reasonable reference for clinical application.Material and method:1.Using silica gel G,chloroform:ethyl acetate:methanol:formic acid?40:5:10:0.2?,cyclohexane:ethyl acetate:acetic acid?15:5:0.4?,n-butanol:acetic acid:water?20:3.5:4?as the agent,the vanillin sulfuric acid solution of 5%ethanol,10%sulfuric acid as chromogenic agent,respectively,on the Shaogan herbs of alcohol extract of paeoniflorin and glycyrrhizic acid,glycyrrhetinic acid were identified by TLC2.HPLC wavelength switching method was used to determine the 6 index components of Paeonia lactiflora Pall.The contents of paeoniflorin,paeoniflorin,liquorice,glycyrrhizin,glycyrrhizic acid and isoglicanin were extracted.3.The fingerprint of alcohol extract of Radix Paeoniae Paeoniae was established by HPLC wavelength switching method4.The HPLC wavelength switching method was used to extract the blood from the abdominal aorta of rats with the extract of ethanol extract,licorice ethanol extract and Paeonia lactiflora extract,and establish the serum fingerprints of alcohol extract,licorice alcohol extract and Paeonia lactiflora.5.The use of diethylstilbestrol and oxytocin Suzao primary dysmenorrhea model rats,intragastric administration of ethanol extract of Radix Paeoniae Alba,Radix Paeoniae Alba and licorice extract on alcohol sweet herbs in rats at different time points of retinal venous plexus blood by HPLC chromatography,simultaneous determination of paeoniflorin and glycyrrhizic acid,licorice acid composition,dynamic behavior of paeoniflorin and glycyrrhizic acid,glycyrrhetinic acid drug.Result:1.In the TLC of the alcohol extract of Radix Paeoniae Alba,the same color spots were displayed in the corresponding position of the paeoniflorin,and the negative solution had no spots.365nm was observed under ultraviolet light.The chromatographic location of paeony and Glycyrrhiza in alcohol extract and glycyrrhetinic acid was identical with orange yellow fluorescence spots,and negative liquid was not found.365nm was observed under ultraviolet lamp.The same color spots were found in the ethanol extract chromatography and the glycyrrhizic acid reference substance,and the negative spots were not found.2.In this experiment,the content of 6 index components was determined by HPLC wavelength switching method.The content of paeoniflorin was 6.904⁷.091 mg/g,and the content of isoglycyrrhizin was slightly lower than 0.2494⁰.2568 mg/g,.This method is stable and feasible,and can be detected at the maximum absorption point.3.The fingerprint of herbal extract of Radix Paeoniae Alba Gan alcohol was established.9 different batches of herbal extracts of Radix Paeoniae Alba sweet alcohol,according to the integrity and the fingerprint of the total,with 22 common peaks,of which 9 peaks from 15 peaks from peony,licorice,has a total of 2 the results show that the peak,the fingerprint of herbal extract of Radix Paeoniae Alba sweet alcohol,the peak area of effective components is large,and other components of the peak area is very small.4.The establishment of peony Gan serum fingerprint of herbal extract on alcohol using HPLC wavelength switching method,the serum containing extracts in baishaogancao alcohols were detected 16 components,including 14 peaks from 8peaks from liquorice,Radix Paeoniae Alba,6 common peaks can be determined,there are 5 ingredients,by contrast the control,the 5 prototype components are albiflorin,paeoniflorin,liquiritin,glycyrrhizic acid,glycyrrhetinic acid.Under the same chromatographic conditions,6 peaks were not detected in the in vitro test solution of Paeonia lactiflora and Glycyrrhiza alcohol extract?or lower than the detection limit?.It is speculated that the 6 components may be metabolites.5.The Tmax of alcoholic extract of Angelica sinensis was only 0.5 h,and the Tmax of ethanol extract of Angelica sinensis was 0.625�0.21 h.The AUC_0-t of oral ethanol extract was 9.239�0.47.The Cmax was 3.044�0.37.The AUC_0-t was 7.751�0.52and the Cmax was 2.609�0.14 for the ethanol extracts of the white peony roots and herbs,and the T_1/2 was 20.83�8.34 h for the ethanol extracts of the white peony.The T_1/2 was 18.71�5.43 h for the alcohol extracts of G.lucidum and glycyrrhizae,and the Tmax of glycyrrhizic acid was 4 h for glycyrrhizic alcohol extracts alone,and the Tmax of g.h,gaccucic acid alone gavage ethanol extract AUC_0-t was 19.27�0.87,Cmax was 2.665�0.23,T_1/2 was 4.704�2.59 h,gavage of Chinese herbal medicine glycyrrhizic acid when gavage AUC_0-t was 11.72�0.60,Cmax was 3.295�0.42,and T_1/2 was 20.56�13.92 h.The glycyrrhetinic acid AUC_0-t was 27.82�0.68,Cmax was2.628�0.26,and T_1/2 was gavage glycyrrhizic alcohol extract alone.The AUC_0-t of glycyrrhetinic acid was 28.73�1.02 h,which was 31.56�1.36,Cmax was 3.064�0.24,and T_1/2 was 5.337�0.36 h.Conclusion:Based on the study of traditional Chinese medicine extract Shaogan science on alcohol,establish the quality standard of Radix Paeoniae Alba herbal extract on GaN alcohol,discusses the pharmacokinetics of herbal extract of Radix Paeoniae Alba sweet alcohol,so that the white Shaogan of herbs extract on alcohol to be scientific,accurate and perfect.A positive exploration was made for the compatibility mechanism of alcohol extracts and the research on the basis of medicinal substances for Radix Paeoniae Paeoniae.
