Purpose: To investigate the effect of scar-moxibustion on the early treatment of rat bilateral Ganshu(BL18)and Zusanli(ST36)acupoints to observe the effects of canonical Wnt signaling pathway on the expression of ?-catenin and secreted frizzled-related protein(s FRP1).To explore the possible mechanism of scar-moxibustion pretreatment in preventing primary hepatocellular carcinoma(HCC)at signal transduction level and to demonstrate the advantages of selecting Back-shu point in preventing primary liver cancer.Material and method: Select SPF grade,12 male Wistar rats were randomly selected as the control group.The remaining 36 rats were randomly divided into model group,pretreated Ganshu(BL18)group and pre-treated Zusanli(ST36)group with 12 rats in each group.Two weeks before the model was made,the two groups were treated with scar moxibustion(0.5 g)in the corresponding ST36 and BL18 acupoints.Rats in the control group and the model group were given the same grasping and fixed stimulation but no scar moxibustion treatment.Two groups of pretreatment groups were intraperitoneally injected with diethylnitrosulfide(50 mg/kg)in the model group,and then a rat primary liver cancer model(PLC)was established after a planned dose of scar moxibustion.The control group had no interventional treatment and was free to drink water and food.During the course of the experiment,net weights were measured in each group of rats,routinely performed once weekly,and body weight gains or losses were recorded.The expression of ?-catenin and s FRP1 were measured by western blot and HE staining was used to observe the pathological changes of the liver.Results: 1.HE staining showed: There groups compares with the control group,the model group,pretreatment BL18 group and pretreatment ST36 group rats.Trabeculae,and liver sinus were disorderly arranged,and accompanied by varying degrees of deep staining.Contrasted with the model group,the number of the pretreatment BL18 group and the pretreatment of ST36 group significantly reduced in term of the number of cancer cells,in which pretreatment BL18 group is best than other groups.2.Rats weight trend: Before ten weeks ago,total weight of body has became increased.From the beginning of 10 weeks,the body weight of rats in the control group was significantly decreased except for the control group,and the model group continued to decline from the 12 th to the 14 th week.On the contrary,the weight of ST36 group and BL18 group both has increased.At 10,12,14 weeks,contrasted with other groups include the control group,ST36 and BL18 group,the body weight of rats significantly reduced.Compared with the control group weight of the rats,the Zusanli(ST36)group and the Ganshu(BL18)group were significantly different(P <0.05).Compared with the pretreated groups,the rats weight of the Zusanli(ST36)group and the Ganshu(BL18)group were significantly different(P <0.05).3.Western Blot Results: Compared with control group,other groups including model group,pretreatment BL18 group and pretreatment ST36 group,three groups,the content of s FRP1,was significantly decreased(P<0.05),.In the contrary,the expression of ?-catenin was significantly increased(P<0.05).Compared with the model group,the expression of s FRP1 was increased in the pretreatment group(P<0.05);the expression of ?-catenin was significantly decreased(P<0.05);the expression of s FRP1 in the pretreated Ganshu(BL18)group was higher than that of the pretreated Zusanli(ST36)group with slightly higher(P<0.05).Conclusions: 1.Scar moxibustion Ganshu(BL18)points and Zusanli(ST36)points can regulate the Wnt signaling pathway expression of ?-catenin and secreted frizzled-related protein(s FRP1)levels,reducing the proliferation of cancer cells effectively.2.Scar moxibustion Ganshu(BL18)points and Zusanli(ST36)points on primary liver cancer can be prevented in some degrees,but the analysis of the differences between the indicators,the effect of Ganshu(BL18)acupuncture point was better than Zusanli(ST36)points.As a result,in this study,scar-moxibustion in Gushu(BL18)is more specific in term of caring primary liver cancer.
