Purpose:In this study,we used diethylnitrosamine(DEN)to induce primary hepatocellular carcinoma(HCC)as a research object.Maize moxa cone was used to perform scar moxibustion pretreatment in Zusanli(ST 36),Ganshu(BL 18).To observe the effect of scar moxibustion moxibustion on the expression of Wnt signal transduction pathway in primary hepatocellular carcinoma(HCC),and to further explore the mechanism of scar moxibustion on primary liver cancer and to clarify some modern mechanisms of scar moxibustion prevention and treatment of hepatocellular carcinoma,For the future scar moxibustion to protect primary liver cancer,delay the development of the disease to provide molecular biology basis.Material and method:A total of 48 Wistar rats were randomly divided into blank group(n = 12),model group(n = 12),pretreatment Zusanli group(n = 12)and pretreatment Group(12).After a week of adaptive feeding,the moxibustion was performed on the pretreated liver and Ganshu and the pretreatment of the Zusanli group.The quality of the moxa was 0.5 g / grain / strong,12 strong,The rats were burned,blank and model rats were not treated with scar moxibustion.After the end of treatment,in addition to the blank group,were used intraperitoneal injection of DEN method to establish rat primary liver cancer model.DEN dosage in accordance with the weight of 50 mg per kilogram,modeling for 14 weeks:1-4 weeks,2 times a week injection;5-14 weeks,once a week injection,injection time in the morning 8:30-10:30.At the end of the model,rats were fasted for one day,10% chloral hydrate anesthesia,abdominal aorta blood collection,centrifugation to take serum for examination,take part of the rat liver tissue with sterile EP tube preservation,liquid nitrogen soaking,-80 ? refrigerator frozen,the rest of the liver tissue with 4% paraformaldehyde fixed,protected from light.Detection of alpha-fetoprotein(AFP)??-glutamyl transpeptidase(?-GT),fucosidase(AFU)content.The expression of Wnt3 a and Wnt10 b m RNA in Wnt signaling pathway was detected by q PCR method.Observation of Pathological Changes of Liver Tissue Structure in Rats by HE Staining.Results:1 HE staining results showed that: compared with the blank group,the model group,pretreatment Ganshu and pretreatment Zusanli rats liver liver trabeculae,liver sinus was disorderly arranged,and accompanied by varying degrees of deep staining Compared with the model group,the number of pretreatment liver and liver group and the pretreatment of Zusanli group significantly reduced the number of cancer cells,which pretreatment Ganshu effect is more significant.2 serological tests: The serum levels of AFP,AFU and ?-GT in the model group,pretreatment Zusanli group and pretreatment Ganshu group were significantly higher than those in the blank group(p<0.05).The serum levels of AFP,(p<0.05);Compared with the model group,the levels of AFP,AFU and ?-GT in the serum of the rats were significantly lower than those in the model group(p<0.05).Compared with the pretreatment Zusanli group,the contents of AFP,AFU and ?-GT in the liver were significantly increased(p<0.05).3 Rt-PCR results show:The expression of Wnt10 b and Wnt3 a m RNA in the liver tissue of the model group,the pretreatment Zusanli group and the pretreatment Ganshu group were significantly higher than those in the blank group(p<0.05),and the difference was statistically significant(p <0.05);Compared with the model group,the m RNA expression of Wnt10 b and Wnt3 a were significantly decreased in the pretreatment Ganshu group and the pretreatment of Zusanli group(p<0.05).Compared with the pretreatment Ganshu group,the expression of related genes in the Zusanli group was significantly higher than that in the pretreatment group(p<0.05).Conclusions:1 Scar moxibustion Gushu points and Zusanli points can regulate the Wnt signaling pathway Wnt10 b,Wnt3a gene expression levels,effectively reduce the proliferation of cancer cells.2 Scar moxibustion Ganshu points and Zusanli points on primary liver cancer have different degrees of prevention,but the analysis of the differences between the indicators,liver shu acupuncture point effect is better than Zusanli points,so this study that scar moxibustion Gushu acupuncture treatment of primary liver cancer is more specific.
