Purpose: The Primary hepatic carcinoma(PHC)model induced by intraperitoneal injection of diethylnitrosamine(DEN)in Wistar male rats was used as the research object.Wheat size moxa were placed in rats Zusanli points and Ganshu points for scarring moxibustion treatment.By observing the morphological changes of the liver,the expression of Wnt signaling pathway-related protein and gene expression in PHC rats after Scar moxibustion.Evaluate the intervention effect of different acupoints of Scar moxibustion on PHC and explore the mechanism of Scar moxibustion on PHC.Material and method: 60 SPF Wistar male rats were randomly selected 12 into blank group.The remaining 48 rats were induced by intraperitoneal injection of DEN solution in a PHC rat model,and the blank group was injected with the same amount of physiological saline in the same manner.Model group,Ganshu group,Zusanli group and doxorubicin group rats were injected intraperitoneally with DEN solution to induce PHC rat model.The modeling period lasted for 14 weeks.The DEN solution was injected intraperitoneally twice a week from the first 1-4 weeks after the model was established,and once a week during the fifth to the 14 th week.After the model was established,48 rats were randomly divided into model group,Ganshu group,Zusanli group and doxorubicin group.Both the Ganshu group and Zusanli group were given a scar-moxibustion intervention at the 15 th week of modeling.At the beginning of the 15 th week of modeling,the rats in Ganshu group and Zusanli group were placed the wheat size moxa after respectively evenly applied with garlic juice,the quality of each grain moxa about 0.5g,the amount of moxibustion at per acupoint is 12.After a burnout with tweezers removed and then replaced,bilateral moxibustion at the same time.Doxorubicin group rats were injected with doxorubicin four times a week starting from the 15 th week of modeling,twice a week.The liver samples were taken from the rats in the 17 th week of modeling.The changes of liver appearance were observed with naked eye.The pathological changes of the liver were observed by HE staining.Western blot was used to detect the expression of s FRP1 and RT-PCR to detect the expression of ?-catenin m RNA.After obtaining the data,the statistics were processed.Results: 1 Appearance of rat liver: In the blank group,the shape of the liver was smooth surface,ruddy color,sharp edge of the livers.The livers of the model group were enlarged,the colors were mixed,the appearance of the liver edge was blunt and multifractal,the nodules were more,the yellow-green tumors appeared in some areas,thick ascites accompanied by bad smell.Livers in Ganshu group rats color reddish brown,slightly rough surface,a small amount of large nodules on the surface of the liver,no obvious tumor.Zusanli group rats liver color reddish brown,rough surface nodules and some slightly larger,the liver margin is not smooth.Doxorubicin group rats liver color reddish,rough surface nodules exist,some large nodules cloudy color and prominent liver surface,liver edge uneven.2 HE staining of liver sections: Compared with the blank group,the trabecular and hepatic sinusoids in the model group,Scar moxibustion liver group,Zusanli group and doxorubicin group rats were disorderly arranged and disorderly.Compared with the model group,the number of cancer nests in liver tissue of Scar moxibustion Ganshu,Scar moxibustion Zusanli and doxorubicin groups were significantly reduced.Compared with the doxorubicin group,Ganshu group,Zusanli group rats' liver cells arranged more orderly,liver cells in the same shape,the nucleus size is uniform,in which the morphology of liver sections of scars moxibustion Ganshu group was closer to the blank group.3 Western blot detection: Compared with the blank group,the expression of s FRP1 in model group,Ganshu group,Zusanli group and doxorubicin group were significantly lower(P<0.05).Compared with the model group,the expression of s FRP1 in the Scar moxibustion Ganshu,Scar moxibustion Zusanli and doxorubicin groups increased significantly(P<0.05).Compared with doxorubicin group,the expression of s FRP1 in Scar moxibustion Ganshu,Scar moxibustion Zusanli and doxorubicin groups increased significantly(P<0.05).Compared with Zusanli group,the expression of s FRP1 in the Scar moxibustion Ganshu increased significantly(P<0.05).4 RT-PCR detection: Compared with the blank group,the expression of ?-catenin m RNA in the model group,Ganshu group,Zusanli group and doxorubicin group were significantly higher(P<0.05).Compared with the model group,the m RNA expression of ?-catenin in the Scar moxibustion Ganshu,Scar moxibustion Zusanli and doxorubicin group were significantly lower(P<0.05).Compared with the doxorubicin group,the m RNA expression levels of ?-catenin in the Scar moxibustion Ganshu and Zusanli groups were significantly lower(P<0.05);Compared with Zusanli the sham group,the m RNA expression of ?-catenin in the Shuyu moxibustion group was significantly lower(P<0.05).Compared with Zusanli group,the m RNA expression of ?-catenin in the Ganshu group was significantly decreased(P <0.05).Conclusion: 1 Scar moxibustion can improve the liver state and pathological morphology of liver tissue of PHC rats to a certain extent.2 Doxorubicin,scar moxibustion at Ganshu acupoint and Zusanli acupoint can up-regulate the expression of s FRP1 and down-regulate the expression of ?-catenin m RNA in liver tissue of PHC rats.3 The effect of scar moxibustion at Ganshu and Zusanli acupoints on PHC is better than that of doxorubicin,and the curative effect of scar moxibustion on Ganshu is more specific than Zusanli.The mechanism may be to delay the onset of PHC by regulating the expression of s FRP1 and ?-catenin m RNA.
