Polygonatum Sibiricum Red. belongs to Liliaceae family, Polygonatum genus, perennial herb. The dried rhizome can be used as medicine with lowering blood pressure, hypoglycemic, regulate immune system, antioxidant, antibacterial and anti-inflammatory activity.This study, we mainly research the chemical constituents of Polygonatum Sibiricum Red. and its potential anti-inflammatory activity. Preliminary experiment which is screening the anti-inflammatory activity of extracted components from PE, TCM, EA, NBA and water. We used LPS-induced RAW264.7 cells as model and NO inhibition rate as index to study the anti-inflammatory activity of each extracted components. The results showed that n-butanol extracted components has the greatest NO inhibition and potential anti-inflammatory activity, then we focus on researching the n-butanol extracted components. Using a variety of chromatograph and semi-preparative HPLC separation methods, we got 32 compounds, then we indentified 27 compounds which according to its physicochemical properties and spectral characteristics as following:Perlolyrin(1),4-(9H-?-carbolin-1-yl)-4-oxo-but- 2-enoic acid methyl ester(2),5-(9H-3-carbolin-1-yl)-pentane-1,2,5-triol(3.4), Flazine (5),2,3,4,6-tetrahydro-lH-(3-carboline-3-carboxylic acid(6),5-hydroxy pyridine-2-carboxylate(7), Polygonapholine A(8), Adenosine(9),5-oxo-pyroli-dine-2-carboxylic acid methyl ester(10),5-oxo-pyrrolidine-2-carboxylic acid butyl ester(11), (±)-5,7-dihydroxy-6,8-dimethyl-3-(2',4'-dihydroxybenzyl)-chroman-4-one(12), bean-5-ene -3?,7a-glycol(13), bean-5-ene -3?,7?-glycol (14), ?-sitosterol(15), ?-daucosterol(16),2-hydroxy-benzoic acid(17), malonic acid dibutyl ester(18), dodeca-4,6,8-trienoicacid-2-hydroxy-1-hydroxymethyl-ethyl ester(19), succinic acid(20), (3-D-fructofuranosyl-a-D-Glucopyranoside (21), methyl-a-D-arabinofuranoside(22), methyl-a-D-fructofuranoside(23), fructose(24), methyl-a-D-galactofuranoside(25), glycerophosphorylcholine (26),1,2-dioleoyl phosphatidylcholine(27), in which compounds 1.2.3.4. 5?6?7?10?11?13?14?18?19?20?26?27 were isolated from Polygonatum genus for the first time and compounds 2?3?4 were isolated from natural origin for the first time. It enriched the chemical constituents and had important taxonomic significance of polygonatum. We monitored objective ino by UPLC-Q-TOF/MS confirming that amino acid derivatives were naturally present in Polygonatum genus rather reaction products.We used LPS-induced RAW264.7 cells as model and NO inhibition rate as index to study the anti-inflammatory activity of each compounds and polysaccharide of Polygonatum. The results found that amino acid derivatives, alkaloids and polysaccharide showed high NO inhibition rate, sterols showed low NO inhibition rate, indicating that amino acid derivatives, alkaloids and polysaccharide may be have good potential anti-inflammatory activity. On one hand, it verified a good anti-inflammatory of Polygonatum, and on the other hand it enriched the material basis of anti-inflammatory of Polygonatum.
