Purpose:The purpose of this study was to investigate the anti-aging effect of gynostemma pentaphyllum seven-leaf bile glycoside XVII(GypenosideXVII,GP-17)on skin photoaging mice.The serum prostaglandin 2(PGE2)in skin photoaging mice was detected by GP-17 The effects of cycloxygenase 2(COX-2)and the expression of MMP-1 in skin tissue were studied to explore the anti-aging effect of GP-17 and the mechanism of oxidative stress in mice exposed to ultraviolet radiationMaterial and method:Sixty 7-week-old male Kunming mice were randomly divided into 6 groups with 10 mice in each group,which were blank control group,UV model group,GP-17 high dose group and GP-17 medium dose group.The low dose group of GP-17 and the positive control group were vitamin E(VE)control group.The skin was fully exposed by removing hair with an area of about 2 X 2cm2 on the back of the mouse.Except for the blank control group,the mice in the five groups were irradiated under 40W ultraviolet lamp(UVA UVB).After the 13th week,the mice appeared lazy,burnout,curled up,rough skin and pig hair sign,the texture thickened and deepened,and the skin became bigger.The skin color began to dark red,indicating that the pathological changes of the skin have been in line with the standards of the photoaging model recorded in the literature,which represents the success of the model.The cumulative intensity of UV is 224.427J/cm2 when the irradiation time is over and the irradiation is terminatedAt the beginning of the 14th week,mice in the blank control group and mice in the UV model group were given intragastrically with a certain amount of saline.GP-17 high,middle and low dose groups and VE control group were given high and low dose gynostemma pentaphyllum seven-leaf bile glycosides by intragastrically.VE olive oil.It lasts two weeks once a dayAt the 16th week,the mice were killed,blood was taken through abdominal aortic,and the skin tissue of the exposed part of the back of the mice was takenResults:1.Expression of PGE-2 in serum of miceThe content of PGE2 in serum of each group was significantly higher than that of blank control group(P<0.01).The content of PGE2 in serum of UV model group was significantly higher than that of blank control group(P<0.01).The content of PGE2 in GP-17 high,middle and low dose groups and VE control group was significantly higher than that in blank control group(P<0.01).Compared with the UV model group,the PGE2 content in the high,middle,low dose GP-17 group and VE control group was significantly lower than that in the VE model group(P<0.01).Compared with the GP-17 high dose group,GP-The content of PGE2 in low dose group and VE control group was significantly higher than that in low dose group(P<0.01)(P<0.05),and the results were statistically significant.2.Expression of COX-2 in serum of miceThe content of COX-2 in serum of each group was significantly higher than that of blank control group(P<0.01).The content of COX-2 in serum of UV model group was significantly higher than that of blank control group(P<0.01).Compared with the blank control group,the COX-2 content in the high,middle,low dose GP-17 group and VE control group was significantly higher than that in the blank control group(P<0.01).Compared with the UV model group,the COX-2 content in the high,middle,low dose GP-17 group and VE control group was significantly lower than that in the VE model group(P<0.01).High dose group with GP-17 Compared with GP-17,the content of COX-2 in low dose group and VE control group was significantly higher than that in VE control group(P<0.01)(P<0.05),and the results were statistically significant.3.The content of MMP-1 in skin tissue of mice in each group.The results showed that compared with the blank control group,the content of MMP-1 in UV model group was higher than that in blank control group,and the difference was statistically significant(P<0.01).The content of MMP-1 in GP-17 high,middle and low dose groups and VE control group was significantly higher than that in blank control group(P<0.01).Compared with the UV model group,the content of MMP-1 in the middle dose group and the VE control group was significantly lower than that in the VE model group(P<0.01).Compared with the high dose group of GP-17,the content of MMP-1 in the middle and low dose groups and VE control group was partial.There was significant difference between the two groups(P<0.01)(P<0.05),and there was significant difference between the two groups(P<0.01)(P<0.05)Conclusion:1.When oxidative stress occurred in mice irradiated with 1.UV,the expression of PGE2 and COX-2 inflammatory mediators was increased,MMP-1 was activated,and extracellular matrix was degraded,resulting in skin photoaging2.The contents of PGE2 and MMP-1 in photoaging mice were higher.GP-17 could inhibit the secretion of MMP-1 and reduce the damage of skin photoaging by effectively reducing the overexpression of PGE2,COX-2 inflammatory factors.
