Objective: To study the expression changes of PI3 k,mTOR,p70S6 k mRNA and gamma interferon in patients with immune tolerance to HBV infection and the effect of kidney-tonifying and detoxification on their expression.Methods:By culturing healthy pregnant women and HBV infected pregnant women with immune tolerance period in vitro,the samples were collected on the 8th day,the IFN-? was detected by ELISA(double antibody sandwich method),the PI3 K,mTOR and p70S6 K mRNA were detected by real-time fluorescence quantitative PCR,and the levels of PI3 K,mTOR,P70S6 K mRNA and IFN-? in the culture supernatant of the two groups were compared.At the 8th day,pDCs of pregnant women in immune tolerance period of HBV infection were cultured and divided into 5 groups,which were intervened by kidney tonifying drug plasma,detoxifying drug plasma,kidney tonifying and detoxifying drug plasma,drug free plasma and the same volume medium with drug containing plasma respectively.After48 h,samples were collected,and IFN-? was detected by ELISA(double antibody sandwich method),PI3 K,mTOR and P70S6 K were detected by real-time fluorescence quantitative PCR MRNA,compare the difference between groups.Result:The expression level of IFN-HBV in pregnant women in the immune tolerance stage of HBV infection was significantly lower than that of normal pregnant women(p<0.05),and the expression levels of PI3 K,mTOR and p70S6 K mRNA in pregnant women in the immune tolerancestage of HBV infection were also significantly lower than that of normal pregnant women(p<0.05).Only by tonifying kidney and detoxifying can the expression levels of pDCs PI3 K,mTOR and p70S6 KmRNA of maternal umbilical cord blood in the immune tolerance stage of HBV infection be better improved(p<0.05),so as to further promote the secretion of IFN-?(p<0.05).Conclusion:HBV infection immune tolerance stage maternal umbilical cord blood pDCs PI3 K,mTOR,p70S6 K mRNA,IFN-virus expression decreased,pDCs function impaired,is HBV makes the immune response of the body weakened,resulting in HBV can not be effectively cleared,one of the important reasons for chronic infection.Kidney detoxification immune tolerance in order to better improve HBV infection period maternal blood pDCs PI3 K,mTOR,p70S6 K mRNA expression level,so as to further promote the secretion of interferons alpha,can partly restore the body's immune function in patients with HBV infection,immune tolerance period,increase assistance antiviral efficacy,for further clinical use provides a good theoretical basis.
