Objective:The expression of SIg A,lymphocyte ?4?7 and its homing receptor MAd CAM-1 in intestinal mucosa of spleen qi deficiency mice were detected.From the perspective of regulating the immune barrier function of intestinal mucosa,this paper attempts to explore the regulatory mechanism of Sijunzi Decoction on the pathophysiology of spleen qi deficiency syndrome,and to study the mechanism of repairing intestinal mucosa immunity with traditional Chinese medicine.Methods:The mice model of spleen deficiency syndrome was made by the method of reducing fatigue and eating incoherence.After the model was made successfully and treated with Sijunzi Decoction for 2 weeks,the general physical signs and body weight of the mice were observed.The weight of thymus,spleen and liver were measured,the SIg A content of intestinal mucosa was detected by enzyme-linked immunosorbent assay,the integrin ? 4 ? 7 content of intestinal mucosa was detected by flow cytometry,and the expression of MAd CAM-1 in intestinal mucosa was qualitatively analyzed by immunohistochemistry.Results:In the first week after modeling,compared with the normal group,the scores of model group and Sijunzi Decoction group decreased significantly(P<0.01);After administration of Sijunzi Decoction,the score of Sijunzi Decoction group was higher than that of spleen deficiency model group(P<0.01);There were significant differences in body weight among different groups and weeks(P<0.01);There was no significant difference in thymus,spleen and liver index between model group and Sijunzi Decoction group(P>0.05);Compared with the normal group,the expression of MAd CAM-1 in intestinal mucosa of the model group was less,and that of Sijunzi Decoction group was acceptable;Compared with the normal group,SIg A content and the percentage of ?4?7 positive lymphocytes in ileum mucosa of model group and Sijunzi Decoction group decreased(P<0.01),after administration,s Ig A content and the percentage of ?4?7 positive lymphocytes in the mucosa of Sijunzi Decoction group were higher thanthose of the model group(P<0.01).Conclusion:1.Through the method of "diarrhea and fatigue + diet loss",the change of general physiological characteristics,body weight and TCM syndrome score of mice indicated that the model was successful,and the longer the duration,the more obvious the performance.After the treatment of Sijunzi Decoction,the general physiological characteristics,body weight and TCM syndrome score of spleen qi deficiency model mice can be improved.2.The expression of MAd CAM-1 in the intestinal mucosa of spleen qi deficiency mice decreased,the number of infiltration and adhesion of ?4?7type immune cells to the vascular endothelium decreased,and the secretion of SIg A decreased.Sijunzi Decoction can enhance the expression of ?4?7and MAd CAM-1,and improve the ability of SIg A expression to protect the intestinal mucosal immune barrier.
