ObjtctiveUlcerative colitis(UC)is a chronic non-specific inflammatory bowel disease that is mainly confined to the mucosa and submucosa.In the past 20 years,the incidence of UC in China has gradually increased,and it has become a common disease in the digestive system in China.The disease is difficult to cure,has a high recurrence rate,and has a poor prognosis.It is a lifelong disease and is disabling.It is a modern refractory disease and a hotspot and a difficult point in the research of Chinese medicine in the field of digestive and gastrointestinal diseases.In this study,male Sprague-Dawley rats were used to observe the intervention effect of different doses of Wenqing Kuiyin Decoction on dextran Sulfate Sodium(DSS)-induced ulcerative colitis in rats,and to explore its effect on TLR4/My The effect of the D88/NF-k B pathway.Method(1)Sixty male Sprague-Dawley SD rats,weighing(200±20)g,were fed for one week before the experiment.They were randomly divided into a blank group,a model group,a mesalazine-positive control group,a high-dose group of Wenqing Kuaiyin,a medium-dose group of Wenqing Kuaiyin,and a low-dose group of Wenqing Kuaiyin,with 10 rats in each group.A rat model of ulcerative colitis was constructed by freely drinking 5% DSS in addition to the blank control group.After 24 hours of modeling,drug intervention was given from day 2 to day 8,and material was taken on day 9.The blank group and the model group were given normal saline,and the mesalazine group was administered with 0.03 g/kg/d.The high-dose,medium-dose and low-dose groups of Wenqing Kuijie drink were 1.2 g/kg/d and0.6 g/kg/ d? 0.3g/kg/d?once a day.(2)The general condition of the experimental animals was observed daily,and the food intake and water consumption were recorded.Body weight ? open field test scores ? and irritability scores were recorded on days 3?5and 7 of the administration.A possible syndrome used to evaluate animal models of UC disease.(3)The disease activity index(DAI)was used,HE staining and histopathological scoring were performed,and the activity level of MPO was measured.Used to evaluate UC animal models.(4)Elisa detected serum p-selectin?IL-6?TNF-?levels,Real time-PCR?Western-blot detection of TLR4?My D88?NF-?Bp65 gene and protein expression.The effect of the drugon the TLR4/My D88/NF-k B pathway was evaluated.Results(1)DSS-induced ulcerative colitis in rats.After freely drinking 5% DSS aqueous solution for 24 hours,the mucous membrane pus and the fecal occult blood were positive in the model group,and the body weight decreased slightly compared with the blank group,suggesting that the model was established.success.(2)The body weight of each group was compared.Compared with the blank group,the body weight of the model group was significantly decreased(P<0.05).The high-dose group of mesalazine and Wenqing Kuaiyin could significantly prevent the weight loss of the rats(P<0.05).The market experiment shows that(3)Compared with the blank group,the DAI score of the model group was significantly increased(P<0.01).Compared with the model group,the mesalazine,Wenqing Kuaijie high and middle dose groups can reduce the DAI score of the rats(P <0.05,P<0.01).HE staining results showed that compared with the model group,mesalazine,Wenqing Kuaijie high and medium dose group can reduce the colonic pathological score of rats(P<0.05,P<0.01).(4)ELISA results showed that compared with the model group,mesalazine,Wenqing Kuaijie high and medium dose group can reduce the expression of MPO,P-selectin,TNF-?and IL-6 in rat serum(P <0.05,P<0.01).(5)WB results showed that compared with the model group,mesalazine,Wenqing Kuaiyin high,medium dose group can significantly inhibit the expression of TLR4,My D88 and NF-?B p65 protein in rat colon tissue(P< 0.05,P < 0.01).(6)QPCR results showed that compared with the model group,mesalazine,Wenqing Kuaiyin high and medium dose group can significantly inhibit the expression of TLR4,My D88 and NF-?B p65 mRNA in rat colon tissue(P<0.05).,P<0.01)Conclusions(1)Wenqing Kuijie Decoction has a good therapeutic effect on DSS-induced ulcerative colitis model,which can inhibit the weight loss and colonic pathological damage of the animal model;it can effectively inhibit the expression of serum pro-inflammatory factors and achieve remission.The role of the condition.(2)Wenqing Kuiyin Decoction has a good therapeutic effect on DSS-induced ulcerative colitis in rats.The therapeutic mechanism may be related to the inhibition of TLR4/My D88/NF-?B signaling pathway.
