Objective: To investigate the effect of Uighur-Xipayi KuiJie'an on NF-κB expression in ulcerative colitis model of rats, establish the platform of RNAi technique; inquire into the sensitive point of ulcerative colitis and functional target of Uighur-Xipayi KuiJie'an. Method: There were five groups: normal group, model group, Xipayi KuiJie'an interfered group, 5-ASA positive control group, NS negative control group; Construct animal model with DNCB and acetic acid; semi-quantitative RT-PCR was used to detect the expression of NF-κB mRNA; Construct pcDNA3.1/CT-GFP-TOPO- NF-κB recombinant eukaryotic expression vector and pSilencer1.0-U6-siRNA-NF-κB interference vector, regulate the expression of NF-κB in COS-7 cells using RNAi by transfection, detect by Western blot and inverted fluorescence microscope, then pick up effective siRNA. Result: In model group, the expression of NF-κB mRNA was higher than normal group; contrast to model group, the expression of NF-κB mRNA in Uighur-Xipayi KuiJie'an interfered group, NS negative control group, 5-ASA positive control group were lower, which all showed the statistic significance(P< 0.05); the expression of NF-κB in Uighur-Xipayi KuiJie'an interfered group and NS negative control group did not show the statistic significance (P>0.05). Regulate the expression of NF-κB in COS-7 cells successfully. Conclusion: Maybe NF-κB was the sensitive point of ulcerative colitis but it was not directly related to the functional target of Xipayi KuiJie'an; regulate the expression of NF-κB in COS-7 cells successfully, establish the platform of RNAi technique; which laid the foundation of whole research.
