Based On Data Mining To Explore The Medication Rules Of Early-onset Ovarian Insufficiency In Classic Chinese Medical Records

Objective:In this study,we collected and sorted out the classic medical cases with exact therapeutic effect of traditional Chinese medicine(TCM)on treating premature ovarian insufficiency(POI).We extracted the first diagnosis prescription,analyzed the experience of classic medical cases in treating POI through data mining,then summarized the diagnosis and treatment thinking,so as to provide more objective statistical basis for the inheritance of TCM and provide more reference for POI treating and scientific research.Method:We searched CNKI,Wanfang database and VIP database with the keywords of’premature ovarian insufficiency’ and ’experience’,et al.and searched the library of Beijing University of traditional Chinese medicine manually.We sorted out the TCM classic medical cases on treating POI,screened the effective cases,and extracted the first diagnosis prescription.With the help of TCM inheritance platform v2.5(TCMISS v2.5),the frequency of drug use,four Qi and five flavor meridian,core drug pair and new prescription were analyzed.Based on the above statistical analysis results,we summarized the characteristics of TCM on POI diagnosis and treatment.Result:After screening,119 classic medical cases,119 prescriptions were included,involving 179 traditional Chinese medicines.The research showed that the common traditional Chinese medicines of POI treatment are Dodder,Cooked Rehmannia Glutinosa,Angelica Sinensis,Licorice.Wolfberry Fruit,Epimedium,Fructus Ligustri,Radix Paeoniae Alba,Salvia Miltiorrhizae and Fructus Dogwood,which are mainly tonic for deficiency,invigorating blood circulation and removing blood stasis,and antipyretic.The syndromes of kidney deficiency,kidney deficiency and liver stagnation were the main syndromes in POI,followed by Yin deficiency and blood dryness syndrome,spleen and kidney deficiency syndrome and kidney deficiency and blood stasis syndrome.The common drugs of different syndromes are different.The common drugs of kidney deficiency syndrome were Dodder,Cooked Rehmannia Glutinatum.Angelica Sinensis,Wolfberry and other kidney-reinforcing drugs.The common drugs of kidney deficiency and liver stagnation syndrome were Dodder,Cooked Rehmannia Glutinosa.Angelica Sinensis,Bupleurum,Cyperus Rotundus,and other drugs for tonifying the kidney and soothing the liver.The common drugs of Yin deficiency and blood dryness syndrome were Liquorice,Fructus Ligustri,Cooked Rehmannia Glutinosa.Cyperus Rotundus,Rhizoma Anemarrhenae,etc.The common drugs of deficiency of spleen and kidney syndrome mainly include Dodder,Poria Cocos,Angelica Sinensis,Atractylodes,Himalayan Teasel Root,etc.The common drugs of kidney deficiency and blood stasis syndrome were Angelica Sinensis,Cooked Rehmannia Glutinosa,Szechuan Lovage Rhizome,Dodder,Safflower,Salvia Miltiorrhiza,etc.According to the four characters,the common drugs were mainly warm and peaceful.According to the five tastes,the common drugs were sweet and bitter.According to the channel distributions,the common drugs were mainly distributed to liver,kidney and spleen channel.Through prescription analysis,the common drug pairs were:(1)Cooked Rehmannia Glutinosa,Dodder(2)Dodder,Angelica(3)Cooked Rehmannia Glutinosa,Angelica(4)Dodder,Wolfberry Fruit(5)Cooked Rehmannia,Dodder,Angelica,etc.Drug association rules were:(1)Cooked Rehmannia Glutinosa,Angelica,Wolfberry Fruit->Dodder(2)Cooked Rehmannia Glutinosa,Wolfberry Fruit->Dodder(3)Angelica,Wolfberry Fruit->Dodder(4)Dodder,Angelica,Wolfberry Fruit->Cooked Rehmannia Glutinosa(5)Cooked Rehmannia Glutinosa,Cyperus Rotundus->Dodder,etc.Through unsupervised entropy hierarchical clustering,four potential new prescriptions were obtained:(1)Radix Codonopsis,Radix Astragali,Mulberry and Lily(2)Cooked Colla Corii Asini,White Lentils,Szechuan Lovage Rhizome,Capillary Wormwood Herb,Polygala tenuifolia Willd,Endothelium Corneum Gigeriae Galli(3)Cooked Rehmannia Glutinosa,human placenta,Angelica,Herba Epimedium,Chinese Rose(4)Fructus Psoraleae,Oyster Shell,Schisandra,Dragon Bone,Trichosanthes kirilowii MaximConclusion:The onset age of POI was mostly over 28 years old,and kidney deficiency syndrome might be the common syndrome in clinic.In terms of drug choice,POI mainly focuses on supplementing deficiency drugs,taking into account clearing deficiency heat,removing blood stasis,addressing both symptoms and root causes.The drugs were mainly warm and mild,the tastes were mainly sweet,the channel distributions mainly belongs to liver and kidney channels.The new formulas of potential drug combination obtained by cluster analysis may have certain theoretical value and representativeness,which could provide reference for clinic and experimental research.
