Objective:To investigate the distribution of rs1179251,rs2227485,rs2227484 and rs2227513 polymorphisms of IL-22 gene in Heiyizhuang children in Guangxi,and to compare the distribution of genotype frequency and allele frequency with that of different ethnic groups.Methods:In this study,115 healthy children in Heiyizhuang of Guangxi were collected as the research objects.Four loci of IL-22 gene rs1179251,rs2227485,rs2227484 and rs2227513 were genotyped by SNa Pshot technique.The population representativeness of the research sample was tested by Hardy Weinberg's law of genetic balance.Finally,the experimental data were analyzed by SPSS22.0 statistical software,and the counting data were expressed by frequency and composition ratio,using Chi-square test or Fisher exact probability method to compare the distribution difference of gene polymorphism of different genders and the distribution difference of genotype and allele frequency between Heiyizhuang healthy children in Guangxi and other ethnic groups.Results:(1)CC,CG and GG genotypes were detected at rs1179251 in the above four loci of IL-22 gene in Heiyizhuang healthy children of Guangxi,and the distribution frequencies were 46.1%,46.1% and 7.8% respectively.Compared with rs1179251 genotype and allele were published in the 1000 Genome project database,the difference between African Nigerians and Europeans was statistically significant(P<0.05);but there was no significant difference compared with Beijing Han population and Japanese(P>0.05).(2)The genotypes of AA,AG and GG were detected at rs2227485,and the distribution frequencies were 24.3%,57.4% and 18.3% respectively.The genotype and allele frequencies of rs2227485 were statistically significant different from Japanese(P<0.05);But there was nosignificantly different from the Beijing Han population,the African Nigerians and the Europeans(P>0.05).(3)The genotypes of AA,AG and GG were detected at rs2227484,the distribution frequencies were 0.9%,18.3% and 80.8% respectively.The genotype and allele frequencies of rs2227484 were statistically significant different from those of Nigerians and Europeans(P<0.05);but there was no statistical significance different from those of Beijing Han population and Japanese(P>0.05).(4)Two genotypes of CT and TT were detected at rs2227513,the frequencies of distribution were 7.8% and 92.2% respectively.The genotype and allele frequencies of rs2227513 were significantly different from those of African Nigerians and Europeans(P<0.05).Compared with Beijing Han population,the difference of genotype distribution frequency was statistically significant(P<0.05),but the difference of allele distribution frequency was not statistically significant(P>0.05).Compared with Japanese,there was no significant difference in genotype and allele frequency(P>0.05).(5)There was no significant difference in the distribution of IL-22 gene rs1179251,rs2227485,rs2227484 and rs2227513 between male and female.Conclusion: There is difference in polymorphisms of Heiyizhuang population in Guangxi and the other races and regions.Objective: To study the relationship between the polymorphisms of IL-22 gene(rs1179251,rs2227485,rs2227484,rs2227513)and asthma in Guangxi Heiyizhuang children,and to investigate the relationship between serum level of IL-22 and asthma.Methods: 121 patients with bronchial asthma and 115 healthy controls were selected as the subjects of this study.The SNa Pshot method was used to genotype the polymorphisms of rs1179251,rs2227485,rs2227484 and rs2227513 of IL-22 gene,SPSS22.0 statistical software was used to analyze the data,Hardy Weinberg's law of genetic balance was used to test population representation.The counting data was expressed in frequency and composition ratio.Logistic regression was used to analyze the correlation between gene polymorphisms and genetic susceptibility of asthma.At the same time,the level of IL-22 in serum was detected by enzyme-linked immunosorbent assay(ELISA).The serum level of IL-22 was expressed by x ± s and the serum level between groups were compared by independent sample t test.All the above statistical analysis,P<0.05 was considered as statistically significant difference.Results:(1)The difference of rs1179251 genotype,allele and dominant model(CG +GG vs CC)between asthma group and healthy group were statistically significant(OR:0.493,95%CI:0.285-0.853,P=0.011;OR:0.659,95%CI:0.437-0.993,P=0.046,OR= 0.524,95%CI:0.321-0.881,P=0.015).After adjusting age and gender,the difference was still statistically significant.(2)The difference of rs2227485 genotype and dominant model between asthma group and healthy control group was statistically significant(OR: 0.451,95%CI:0.249-0.815,P=0.008;AG+GG vs AA:OR=0.489,95%CI:0.279-0.857,P=0.012).After adjusting gender and age,the difference was still statistically significant.(3)The genotypes and alleles of rs2227513 were significantly different between asthma group and healthy control group(OR=0.198,95%CI:0.042-0.937,P=0.041;OR=0.205,95%CI:0.044-0.957,P=0.044).After age and gender correction,the difference was still statistically significant.(4)The four loci of IL-22,rs1179251,rs2227485,rs2227484 and rs2227513 had linkage disequilibrium.(5)The result of haplotype analysis showed that there was a significant difference in the distribution of G-G-G-T haplotype between asthma group and healthy control group(OR:0.648,95%CI:0.426-0.985,P=0.041).(6)The average serum IL-22 level in the asthma group was significantly higher than that in the healthy control group,and the difference was statistically significant(P<0.001).There was no statistically significant difference in the serum IL-22 levels between children with CC genotype rs1179251 and those with CG/GG genotype in the asthma group(P=0.674).Conclusion:(1)The polymorphisms of rs1179251,rs2227585 and rs2227513 of IL-22 gene may be related to the susceptibility to asthma in Heiyizhuang children in Guangxi,and rs1179251(CG genotype,G allele and dominant model),rs2227485(AG genotype and dominant model)and rs2227513(CT genotype and C allele)may reduce the risk of asthma.(2)The four loci of IL-22 gene,rs1179251,rs2227485,rs2227484 and rs2227513 have linkage disequilibrium.G-G-G-T haplotype may reduce the risk of asthma.(3)The increased level of serum IL-22 may be related to the incidence of asthma in Heiyizhuang children in Guangxi.
