Objective: To investigate the effect of endurance and resistance exercise on NLRP3 inflammasomes and IL-1? in type 2 diabetic rats induced by intraperitoneal injection of high glucose and high fat combined with streptozotocin(STZ)and its possible mechanism.METHODS: Male Sprague-Dawley rats were randomly divided into normal control group(p C)and diabetic model group(p D),fed with normal feed and high-sugar and high-fat diet respectively.After feeding for 7 weeks,p D group was given a 30 mg/kg chain.Urozotocin was intraperitoneally injected.After 72 hours,non-fasting blood glucose ?16.7 mmol/L was selected into the diabetic group and randomly divided into diabetic model group(D),diabetic endurance exercise group(DA)and diabetes resistance exercise.Group(DR);p C rats were randomly divided into blank control group(C),endurance exercise group(CA)and resistance exercise group(CR).CA,DA,CR,and DR performed non-weight-bearing treadmill exercise and weight-bearing ladder training,respectively.After 8-week exercise intervention,body weight,serum lipids,fasting plasma glucose(FPG),glucose tolerance(AUC),insulin(FINS)and insulin resistance index(HOMA-IRI)were measured.The left ventricular myocardial structure was observed under light and electron microscope.Left ventricular catheterization was used to measure cardiac hemodynamic parameters;ELISA was used to measure myocardial NLRP3,ASC,caspase-1,and IL-1?.RESULTS: After 1.7 weeks of high glucose and high fat combined with one-time intraperitoneal injection of STZ,the model-forming rate of type 2 diabetic rats was 72.2%.2.Compared with group C,myocardium in D group was disordered,some muscle fibers were lysed and ruptured,and mitochondria were swollen and degenerated;body weight and FINS were significantly decreased;FPG,HOMA-IRI,AUC,triglyceride(TG),total cholesterol(CHOL)),Low-density lipoprotein(LDL),Very low-density lipoprotein(VLDL)increased significantly;left ventricular end-diastolic pressure(LVEDP),left ventricular diastolic pressure(LVDP),left ventricular relaxation time constant(Tau)increased significantly,left The maximum rate of decline in intraventricular pressure(-dp/dt max)decreased significantly;myocardial NLRP3,ASC,caspase-1,and IL-1? increased significantly.Compared with group D,the myocardial ultrastructure of DA group was improved,but there was no obvious change in DR group;FPG,HOMA-IRI,AUC,TG,CHOL,LDL,and VLDL in DA group and DR group were significantly decreased,and body weight and FINS were significantly decreased.Increase;LVDP,LVEDP and Tau in DA group decreased significantly,-dp/dt max increased significantly,no significant changes in the above indicators in DR group;DA group NLRP3,ASC,caspase-1 and IL-1? significantly reduced,DR group is not obvious difference.CONCLUSION: The 1.7-week high-sugar high-fat feeding combined with low-dose single-dose STZ intraperitoneal injection can successfully establish a T2 DM rat model.Both 2.8-week endurance and resistance exercise can improve myocardial glucose and lipid metabolism abnormalities in diabetic rats.Endurance exercise is more effective in regulating blood lipids,and resistance exercise is more effective in regulating blood glucose.3.8 weeks of endurance exercise can reduce the activation of NLRP3 inflammasome and IL-1? in myocardium and reduce the inflammatory response of myocardium,thus improving myocardial injury in diabetic rats.Eight weeks of resistance exercise had no significant effect on NLRP3 inflammasome and IL-1?,but no significant changes in myocardial structure and left ventricular diastolic function.
