Previous studies have shown that functional lipids containing conjugated linolenic acid(CLn A)have many effects such as anti-inflammation and anti-oxidation.In this study,the changes of body weight,blood lipid and glucose level were studied in mice after the intake of different proportions of catalpa seed oil and pomegranate seed oil.The mechanism of glucose and lipid metabolism regulation was examined,and the changes of the intestinal flora were analyzed.This study provides theoretical evidence for the regulation of glucose and lipid metabolism in mice by functional lipids.The male mice were randomly divided into five groups.The mice fed with 20% high-fat diet were used as the control group.After 7 weeks of feeding,the growth index of the mice was observed.It was found that both catalpa seed oil and pomegranate seed oil could decrease the body weight growth rate of the mice,while no significant effect on the tail length of the mice was detected.The body mass index(BMI)decreased after catalpa seed oil and pomegranate seed oil intake.Compared with the control group,T-CHO decreased by 42.0% and 31.7%,TG decreased by 23.8% and 39.6%,respectively,after adding 5% and 10% catalpa seed oil,and no significant change in LDL-C content were observed;The content of T-CHO,TG,HDL-C and LDL-C in 5% pomegranate seed oil group were not changed significantly,while the content of TG and HDL-C could be regulated with 10% pomegranate seed oil in serum.IPGTT glucose tolerance test results showed the addition of 5% and 10% catalpa seed oil could significantly improve glucose tolerance in mice.At the same time,glucose tolerance capacity could be improved significantly with 10% pomegranate seed oil,while no obvious effect was detected in 5% pomegranate seed oil group.The results of hematoxylin-eosin(HE)staining showed that the steatosis of liver tissue of mice could be alleviated with catalpa seed oil and pomegranate seed oil,but no histological changes on the intestine were detected.In order to further explore the mechanism of catalpa seed oil and pomegranate seed oil affecting on glucose and lipid metabolism in liver,Western blot results showed that the protein expression of PPAR? could be significantly increased in mice liver with both 10%catalpa seed oil and 10% pomegranate seed oil,further real-time quantitative PCR analysis was performed and it showed that both catalpa seed oil and pomegranate seed oil can up-regulate the m RNA level of PPAR?.The results suggest that the effects of catalpa seed oil and pomegranate seed oil on glucose and lipid metabolism may be related to the expression of PPAR?.16 Sr RNA high-throughput sequencing was used to analyze the intestinal microflora structure of mice.It was found that the ratio of relative abundance of Bacteroidetes to Firmicutes decreased in the catalpa seed oil group,while that of the pomegranate seed oil group increased to a certain extent.At the same time,the relative abundance of ?-Proteus in the experimental group was lower than control group.In summary,we found that body mass index(BMI)and serum levels of T-CHO could be decreased,glucose tolerance and TG and LDL-C,HDL-C level was increased with catalpa seed oil and pomegranate seed oil.The effects of these two oils on glucose and lipid metabolism may be related to the regulation of PPAR? expression.Whether the changes of intestinal microflora structure were involved in the effects of these two functional oils on glucose and lipid metabolism remains to be further studied.
