Olive oil is an edible oil with rich nutritional value.The presence of bioactive components in olive oil such as squalene,polyphenols,and sterols has an important impact on the flavor,stability,and active function of olive oil,but it is currently difficult to accurately and quickly detect these bioactive components in olive oil.In this paper,olive oil is used as a raw material to establish a rapid detection method for the content of squalene,polyphenols and sterols in olive oil,and on this basis,the optimization of pretreatment conditions is analyzed and discussed.The results show that the method can not only simplify the processing steps,process samples in batches quickly,use less reagents and save resources,but also has low detection limit,good recovery rate and high accuracy,which can meet the biologically active ingredients in olive oil.Simple and rapid detection of squalene,polyphenols and sterols.The main research contents and results are as follows:(1)The method of ultra-high performance liquid chromatography ultraviolet detector for analyzing squalene in olive oil was studied and the operating conditions were optimized;The silica gel solid phase extraction purification column was selected,and the ACQUITY UPLC BEH C18 column was used for analysis at 30?The detection limit of squalene is 0.367 mg·kg-1,and the recovery rate is 100.836%,which meets the requirements for rapid and accurate detection of squalene in olive oil.(2)The method of ultra-high performance liquid chromatography fluorescence detector for analyzing hydroxytyrosol,tyrosol and oleuropein in olive oil was studied and the operating conditions were optimized;Polyamide solid phase extraction purification column was selected at 30?,the ACQUITY UPLC BEH C18 column can be used to achieve good separation of the three polyphenols.The detection limits of hydroxytyrosol,tyrosol and oleuropein are 0.053,0.098 and 0.069 mg?kg-1,and the recovery rates are 91.493%,91.563%and 92.244%,respectively,meetting the requirements of accurate and rapid detection of three polyphenols in olive oil.(3)The method of ultra-high performance liquid chromatography ultraviolet detector to analyze rapeseed sterol,campesterol,stigmasterol and?-sitosterol in olive oil was studied and the operating conditions were optimized;The amino solid phase extraction purification column was selected at 30?,the ACQUITY UPLC BEH C18column can be used to achieve good separation of the four sterols.The detection limits of rapeseed sterol,campesterol,stigmasterol,and?-sitosterol are 0.174,0.295,0.160,and 0.157 mg?kg-1,respectively.The recovery rates are 84.728%,87.516%,86.295%,88.674%,which can meet the fast and accurate detection of four sterols in olive oil.
