In this study,proanthocyanidins were extracted from Chardonnay grape seeds by solvent extraction.oligomeric proanthocyanidins and polymeric proanthocyanidins were obtained by rapid separation of polymerization degree.The polymeric proanthocyanidins were degraded by polymerization degree.The changes of components under different degradation conditions were analyzed by HPLC;The antioxidant activity of procyanidins in vitro was studied by conventional chemical method and cell test,and the anti-aging effect of procyanidins was compared by establishing D-galactose-induced aging mouse model:1.The solvent extraction method was used to extract proanthocyanidins.Through single factor and response surface optimization design,the optimal extraction conditions were determined as follows:ethanol concentration 60%,solid-liquid ratio 1:6 g/m L,extraction time 68 min,extraction temperature50?,under which the extraction rate of proanthocyanidins was 3.79%.2.The degree of polymerization of polymer proanthocyanidins degraded by sodium hydroxide was studied and optimized by response surface methodology.Under the optimal conditions of sodium hydroxide concentration of 4.40%,treatment time of 31 min and treatment temperature of 59?,the degree of polymerization of proanthocyanidins was 2.37.The results showed that the peak areas of monomer,epicatechin gallate(ECG)and dimer increased by 1.36,27.37 and 2.48 times respectively,and the total peak area increased by 1.80 times.3.The antioxidant activity of proanthocyanidins was studied by the combination of conventional chemical method and cell test.The results showed that the antioxidant activity of procyanidins with different polymerization degrees was positively correlated with the concentration within a certain concentration range.The results showed that the degradation of sodium hydroxide could improve its antioxidant activity,and the DPPH and ABTS radical scavenging ability of the treatment group rich in procyanidin dimer and ECG was stronger.Use 10?M?amyloid(A?)to explore the antioxidant activity of oligomeric and high polymeric proanthocyanidins,PC12 cells were treated with(A?1-42).The results showed that:0.1,0.2,0.5,1.0,2.0,5.0 and 10.0?g/m L of oligomeric proanthocyanidins and 0.5,1.0?g/m L high poly proanthocyanidins could inhibit A?1-42significantly increased the survival rate of PC12cells(p<0.01).4.The anti-aging effect of procyanidins was further studied by establishing D-galactose aging mice model.The results showed that the contents of immunoglobulin G(Ig G)in oligomeric procyanidins group and oligomeric procyanidins group were significantly lower than those in model group,and the contents of interleukin-1?(IL-1?)and interleukin-6(IL-6)in oligomeric procyanidins group were significantly lower than those in oligomeric procyanidins group The results of HE staining showed that the degree of liver and kidney injury was improved after treatment with oligomeric procyanidins,but the effect of oligomeric procyanidins was not obvious.
