| Gynostemma pentaphyllam is warm and bitter in green soup.Although tasting bitter in mouth,sweetness appears when flowing through throat.Because of high nutritional value,it is known as"Southern Ginseng".Chokeberry,"King of Berries",contains many healthy components but the forbidding sourness hardly satisfies consumers.The first part of this experiment is the production of Gynostemma pentaphyllam-chokeberry beverage.The ratio of composite sweeteners(xylitol,erythritol and mogrosides)is optimized by response surface method with the sensory evaluation as the response value.After obtaining the optimal sweeteners,the response surface method is also used for optimizing the beverage formula(tea,chokeberry,sweetener,citric acid).The beverage can then be produced by homogenizing,desecrating,filling,sterilizing and cooling.The second part is to test the physical indexes,chemical indexes and microbial indexes of the beverage.The third part is to find the free radical scavenging capacity of beverage in different concentrations in vitro,the antioxidant capacity in CAA and the obesity regulating ability of it.There are several specific conclusions in this experiment.The response surface is used to optimize the compound sweeteners.Xylitol,erythritol and mogrosides are 3.17%,3.89%and 0.02%respectively,with the predicted score 95.48.The actual amount of them are modified to 3.2%,3.9%,0.02%separately for easier production.Sensory evaluators are selected to score the modified sweeteners in score 95,which is consistent with the predicted results.The response surface method is used to optimize the beverage formula.The amount of tea,chokeberry juice,citric acid and sweeteners are 32.56%,4.07%,0.14%and 6.77%respectively,with the predicted score 97.12.The actual amount of them are modified to32.6%,4.1%,0.14%and 6.77%separately for easier production.Sensory evaluators are selected to score the modified beverage formula in score 97,which is consistent with the predicted results.The brewed tea and the recovered chokeberry juice are mixed with citric acid and sweeteners according to formula.0.05%sodium alginate stabilizer is then added.Homogenization,deviation,sterilization and other processes are also needed.The beverage is in purplish red.It is clear without precipitation,moderate sugar-acid ration as well as multiple aromas.The pH value of the beverage is 3.54.The soluble solid is 6.47%.7.31 g/kg total acid,3.66 g total sugar and 1.73 g reducing sugar are in per 100 m L beverage.Besides,there are14.08 mg/m L total phenol,12.4 mg/100 m L total anthocyanin,0.8 g/100 m L total saponin as well as 51.8 mg/100 m L total flavone.Sweetness is the main flavor while bitterness is the weakest in the beverage.The total bacterial colony,fungal colony and yeast colony of the beverage is 52 CFU/m L,6 CFU/m L and 6 CFU/m L respectively.No Escherichia coli is detected.The beverage scavenging rate of DPPH,ABTS~+,peroxygen free radical is 87.53±2.38%,91.20%±2.56%,292.44±21μmol Trolox/100 g,respectively.Besides,CAA method is also used.The free radical scavenging ability of the beverage after PBS cleaning is 218.64±24μmol Q/100 g and 570.12±34μmol Q/100 g without PBS washing.Chitosan is regarded as the control group.Mice are given beverage,chitosan and purified1%saline for 6 consecutive weeks.After 6 weeks,the body weight of the model group,control group and experimental group are 27.85±2.68 g,24.05±1.42 g and 23.45±1.07 g,respectively.In animal body composition analysis,anatomy and tissue mass weighing,it can be seen that the epididymal fat mass of mice in the model group is the largest,significantly lower than the control group.Compared with the control group and the experimental group,the levels of TG,TC,ALT and AST decreased.Besides,the effect of the beverage in inhibiting obesity is better than chitosan.In conclusion,there is no functional drink on the market that uses Gynostemma pentaphyllam and chokeberry as raw materials.This research provides a new method for use of G.and chokeberry and a corresponding theoretical basis of the functional beverages by enterprises. |
