Metal-organic compounds,such as metal-organic framework(MOF)and infinite coordination polymers,have attracted more attention for scientists.These kinds of nanomaterials have excellent electrochemical performance.Therefore,it has a broad application prospect in the field of electrochemistry and luminescence chemistry detection.In this thesis,based on the composite of metal-organic compounds and other materials,the biosensors with excellent performance were constructed and applied tothe detection of target objects.The maincontents areas follows:In Chapter 1,the classification of nanocomposite materials,biosensors and their applications and then the principle of fluorescence analysis are described briefly.Finally,the main content and significanceofthis thesis was pointed out.In Chapter 2,an electrochemiluminescence(ECL)sensor was synthesized based on ZIF-8modified with multilayer Ti3C2 TX nanomaterials and then used to detect HIV-1 protein.The linear relationship ?I = 0.9528 lgc–7.887(R2 = 0.9935)was obtained in the range from 1 f M to 1 n M with the detection limit(LOD)of 0.3 f M.The results showed that the biosensor not only improved the electrical conductivity greatly of the electrode,but also had excellent electrochemical performance.Inaddition,satisfactory results wereobtained for the detection of HIV-1 protein inactual samples.In Chapter 3,the cyclodextrin based MOF(CD-MOF)was synthesized and then used to encapsulate Ru(bpy)32+ to construct a sandwich-type ECL biosensor.Under the optimal experimental conditions,the established biosensor has a high sensitivity with a detection limit of 6 pg m L-1 and a linear range from 0.1 to 200 ng m L-1 for the detection of CYFRA21-1.After biotoxicity test,it can be seen that CD-MOF@Ru(bpy)32+ has low toxicity to living cells.Finally,the designed biosensor was further applied to the determination of CYFRA21-1 from A549 lung cancer cells of mouse with the satisfactory recoveries.In Chapter 4,Cu nanoclusters(Cu NCs)were wrapped by infinite coordination polymer TB-GMP with frame structure to form a molecular fluorescent biosensor with excellent luminescence properties.The linear relationship between the difference of fluorescence intensity and the alkaline phosphatase concentration during the range of 0 ~ 2 U m L-1 was I/I0 = 0.2774 c +0.01370(R2 = 0.9934),and the LOD was 0.02 U m L-1.The synthesized Cu NCs@Tb-GMP nanocomposite was further applied to the detection of alkaline phosphatase in lake water with the recoveryof 96.7 ~ 108.2% was obtained.
