With the rapid development of agriculture in China,animal husbandry,fishery and other industries have produced a large number of waste by-products,such as skin,bone,viscera,etc.Natural collagen extraction of these waste can not only make agricultural waste more valuable utilization,but also improve the local environment.In the field of daily use chemicals,collagen and its degradation products have become one of the most important applications of collagen products due to the wide variety of products,wide audience,and relatively wide requirements on the performance of collagen products.However,in practical application,the differential comparison and analysis of structure and properties of different types and sources of collagen or collagen hydrolysates are not perfect,and the influence of different sources and types of collagen on the function of daily chemicals is not clear,which limits the scientific application of various collagen products to a certain extent.For this reason,10 kinds of collagen products with different sources and different structures were selected in this paper to systematically analyze and compare their physical and chemical properties.On this basis,collagen essence was prepared to discuss the advantages and disadvantages of different types of collagen products in the aspects of moisturizing,soothing and repairing efficacy.The main research contents are as follows:1.Obtained high quality natural collagen of turbot fish by self-lifting in the laboratory.Deep-grinding collagen fragments and heat-denatured collagen peptides were prepared from natural bovine calcaneus collagen(purchased)and natural turbo fish collagen(self-extracted).A total of 10 kinds of collagen products,such as collagen peptides and recombinant collagen purchased from the market,were taken as the analysis objects to conduct a comprehensive analysis of their physical and chemical properties.The results of amino acid composition and differential scanning calorimetry showed that the thermal stability of natural collagen was closely related to its amino acid composition and habitat environment.The higher the degree of amino acid hydroxylation,the better the thermal stability.The thermal shrinkage temperature of natural collagen was bovine Achilles tendon>.Of turbot.The results of spectroscopic analysis showed that,except for the absence of collagen ultraviolet and infrared characteristic peaks,the other 9 kinds of recombinant human collagen had characteristic absorption peaks of collagen.The results of circular dichrochromatographic analysis showed that the natural active collagen and the frozen ground collagen fragments had a three-stranded helix structure,and had relatively complete molecular structure activity,but the other collagen products lost the three-stranded helix structure,and presented a random curl conformation.The results of gel electrophoresis showed that the molecular weight of natural active collagen was large and concentrated,the molecular weight of freeze-grinding collagen fragment was small and wide,the molecular weight of heat-denatured collagen was small and concentrated,and the molecular weight of collagen peptide was small.The results of solubility and viscosity analysis showed that the solubility of collagen products was directly related to its molecular weight,and the solubility of collagen peptide was the best,while the viscosity of natural active collagen was the highest.2.The evaluation results of the moisturizing efficacy of the product showed that,at the same molecular weight level,the more the content of moisturizing amino acids,the stronger the moisturizing ability of collagen;At different molecular weight levels,the smaller the molecular weight,the stronger the hydrating ability of collagen and its degradation products.The results of in vitro test showed that the active collagen with greater viscosity had a smaller water loss rate within 2 hours,while the collagen peptide with smaller molecular weight had the weakest water loss and the strongest moisture retention ability after 2 hours.Body surface test showed that collagen peptide products had better moisturizing effect than active collagen products.The results of the evaluation of the restorative efficacy of the products showed that the RBC hemolysis rate of the essence prepared by collagen and its degradation products was very low,among which the stimulation of active collagen was the least.After adding stimulants,the hemolysis rate of active collagen was the lowest,which indicated that the synergistic effect of active collagen and anti-sensitive active substance was the best,and it had a good protective effect on RBC cell membrane.All the products of the moisturizing and repairing essence have good sensory experience,with high stability of p H,normality,hot and cold alternation,and no precipitation,color and odor change,which provides a feasibility basis for product marketization.
