Rice is widely planted in China with rich resources and high yield.Rice bran is the main by-product in the process of rice processing.However,the comprehensive utilization rate of rice bran is less than 20%,and the degree of product development is low.Most of rice bran is used as feed,resulting in a great waste of this renewable resource.Rice bran polysaccharide is one of the most valuable functional active ingredients in rice bran.It has many physiological activities,such as anti-tumor,anti-oxidation,enhancing immunity,reducing blood sugar and blood lipid.In this paper,defatted rice bran is used as raw material to extract rice bran polysaccharide,optimize the extraction process,detect and analyze rice bran polysaccharide,and prepare rice bran polysaccharide based carrier to contain curcumin.The research of this subject is for the industrial production and high value application of rice bran polysaccharide,scientific,reasonable and effective development and utilization of food,medicine and health food with rice bran as raw material,It is of great significance to increase the added value of the whole industry?In this study,defatted rice bran was used as raw material to extract polysaccharides from rice bran by extrusion and ultrasonic assisted enzyme extraction.Orthogonal experiment and response surface method were used to extract polysaccharides from defatted rice bran by extrusion and ultrasonic respectively.The optimal extraction process was as follows: the amount of water added to the material was 25%,the extrusion was carried out at 120 ? and the rotation speed was 150 r/min to obtain extruded defatted rice bran,Then,under the conditions of solid-liquid ratio of 1:20 g/ml,temperature of 71?,ultrasonic power of 400 W,ultrasonic treatment for 72 min,the extraction rate of rice bran polysaccharide can reach 5.11%.The results showed that the rice bran polysaccharide had good water solubility,almost insoluble in ethanol,ether,ethyl acetate,n-butanol and other organic reagents.There were sugars,but no reducing sugar,starch,amino acids and uronic acid.The purity of the polysaccharide was high,and its molecular weight was between5?426 k Da.It contained glucose,galactose,xylose,fructose and other four kinds of monosaccharides.Finally,the infrared absorption spectrum showed that the polysaccharide existed ?-Heteroglycans with glycosidic bonds.Delivery system has been one of the hot spots in related fields because of its characteristics of drug controlled release,targeting,completeness and effectiveness.Carrier material is the key technology to achieve drug controlled release,and polysaccharide natural polymer not only has the characteristics of safety,non-toxic,stability,hydrophilicity and biodegradability,but also has rich resources and low processing cost,so it has good application value as drug carrier material.The purified rice bran polysaccharide was modified with polylactic acid and loaded with curcumin to construct a rice bran polysaccharide based drug loading system.The average effective particle size of the carrier was 213.9 nm,the PDI was about 0.2,the electric potential was-25.3 m V,and the average drug loading was 6.5%.The carrier was evenly distributed and the solution was stable,which indicated that rice bran polysaccharide could be used as a good carrier material.Further optimization of rice bran polysaccharide carrier can not only contain curcumin,but also other fat soluble substances,which can be used as a widely used carrier,and has great development space and application value.
