Bamboo shoots of Phyllostachys praecox are tender and white,delicious in taste and rich in nutrition.Its rich dietary fiber can promote intestinal peristalsis,prevent constipation,but also effectively inhibit the occurrence of diabetes,cardiovascular and brain diseases.As a pure natural pollution-free traditional forest vegetable,bamboo shoots of phyllostachys praecox are deeply loved by the majority of consumers in today ' s society.However,the perennial application of large amount of chemical fertilizer in the growing process of phyllostachys praecox shoot caused serious damage to the ecological environment of the bamboo forest,and reduced the output and quality of bamboo shoots.In this study,the bamboo shoots of Phyllostachys praecox were taken as the test object,under the action of Bacillus subtilis,the changes of bamboo shoot quality and comprehensive evaluation,the metabolism of dietary fiber and the expression of related key genes,and the changes of soil nutrient content were studied from physiological,biochemical and molecular aspects,the effects of Bacillus subtilis on the quality and metabolism of dietary fiber of phyllostachys praecox shoot were studied.The main findings are as follows:1.There are certain differences in the content of each quality index of Lei bamboo shoots with different treatments.Among them,reducing sugar(7.99?30.81mg/g),soluble sugar(34.76?58.82mg/g),dietary fiber(35.22? 39.64%),total phenols(0.11?0.24/g),flavonoids(0.09?0.22/g)content difference is big,the water content(89.15?91.52%),edible rate(59.52?65.86%)difference is not big;Through principal component analysis and cluster analysis,the ranking of Lei bamboo shoots with 10 different treatments was T6,T1,T5,T7,T2,T4,T8,T9,CK,T3.2.The study of dietary fiber metabolism and related enzymes of bamboo shoots found that with the growth of bamboo shoots,the content of DF,IDF,cellulose,hemicellulose and related enzyme activities gradually increased.Compared with T6 treated with Bacillus subtilis,CK has higher content of total dietary fiber,cellulose and hemicellulose with the growth of bamboo shoots,and stronger PAL and POD activities.When the height of the bamboo shoots is 20 cm,they are both significant.Higher than CK.3.The rhizosphere soil environment and bamboo shoot growth of Lei bamboo shoots were studied,and the results showed that the daily growth of Lei bamboo shoots under the two treatments showed no significant difference(P>0.05);the total nitrogen and alkalinity of the soil under the T6 treatment Nitrogen solution content was significantly higher than CK treatment(P<0.05),available phosphorus and available potassium content were significantly lower than CK treatment,there was no significant difference in organic matter content between the two;the number of bacteria in T6 treatment was significantly higher than that of CK treatment,the number of fungi and the release line The changes in the number of bacteria showed no significant difference.4.Transcriptome sequencing of the two treatments of Lei bamboo shoots revealed that a total of 90,441 genes were involved in the expression.The number of differential genes was6563,accounting for 7.26% of the total expressed genes,of which 3883 genes were up-regulated and 2680 genes were down-regulated.The GO function annotation of the differentially expressed genes of Lei bamboo shoots found that the number of organic ring compound binding and heterocyclic compound binding enrichment in the molecular function classification was the largest,the numbers were 1544 and 1541,respectively.The DNA metabolism process and DNA integration enrichment in the biological process classification The number of mitochondria and nuclei is the largest among the cellular components.The classification of KEGG metabolic pathways and significant enrichment analysis found that the differential gene expressions of purine metabolism and plant hormone signal transduction pathways were relatively large,77(11.03%)and 68(9.74%),respectively.Dietary fiber metabolism-related enzyme genes such as PAL,POD,CAD,etc.are up-regulated at the transcription level.
