Rose(Rosa rugosa),also known as red rose,penetrating rose,wandering flower,etc.,is a deciduous shrub of Rosaceae,sweet in nature,warm in taste,and rich in nutrients.This topic useed water and 70%ethanol two solvents to extract the total crude flavonoids in roses.The rose flavonoids extracted from the two solvents were studied for anti-oxidation,anti-aging,anti-tumor,anti-bacterial,and intestinal flora balance.And its anti-oxidant and anti-bacterial physiological activities were applied to food preservation.(1)Study on the extraction conditions and content of total flavonoids from roses.The extraction conditions used in the experiment are 1:10 material-to-liquid ratio leaching in a water bath at 40℃for 2 h.After freeze-drying,the extraction rate of crude rose flavonoids extracted with water(RFW)was 24.70%,and the crude rose flavonoids extracted with 70%ethanol(RFE)was 28.34%.The content of flavonoids in RFW was 47.82±0.152 mg RE/g,and the RFE was 79.08±0.195 mg RE/g.(2)Study on the antioxidant activity of total flavonoids from roses.Rutin was used as a positive control to evaluated the total reducibility of RFW and RFE,DPPH free radical scavenging rate,ABTS free radical scavenging rate and superoxide anion free radical scavenging ability.The IC50of RFW and RFE to ABTS were 0.018 mg/m L and 0.017 mg/m L respectively,and the difference between the two was relatively small.The IC50of RFW and RFE to DPPH were 0.033 mg/m L and 0.021 mg/m L respectively.The IC50values of RFW and RFE for superoxide anion(O2-)were 0.100 mg/m L and 0.094 mg/m L respectively.In general,the overall scavenging ability of RFW and RFE for the three free radicals was ABTS>DPPH>O2-,and the scavenging ability of RFE was better than that of RFW.(3)Study on the anti-aging activity of total flavonoids of roses.This study used C.elegans to establish an anti-aging biological model.Through the life cycle of C.elegans,head swing frequency,body stretching frequency,pharyngeal pump rate,excretion cycle,oxidative stress,and antioxidant enzyme activity in the nematode to determine the anti-aging activity of rose flavonoids against C.elegans.The results showed that 2.0 mg/m L was the optimal concentration of RFW and RFE that can prolong the lifespan of the nematodes,both can extend the lifespan of the nematodes to 20 d,and the half-lethal days were 12.64 d and 10.44d respectively;the oxidative stress results showed the RFE of 2 mg/m L and 4 mg/m L can extend the life of nematodes to 40 h;the RFW of 2 mg/m L and 4 mg/m L can extend the life of nematodes to 36 h.Antioxidant enzyme activity test shows that RFW and RFE can increase the activity of SOD and CAT in C.elegans,thereby exerting anti-aging effects.(4)Study on the anti-tumor activity of total flavonoids of roses.Human hepatocarcinoma cell lines HepG2 was cultured,and methotrexate was used as a positive control.RFW and RFE were applied to HepG2 cells.The inhibition rate was measured by MTT method,the cell morphology,cell migration rate,invasion rate,cell apoptosis and protein expression were measured.The results showed that low concentration rose flavonoids(10-40μg/m L)can inhibit the migration and metastasis of HepG2 cells;high concentration rose flavonoids(40-160μg/m L)up-regulated the expression of pro-apoptotic proteins p53 and Bax,and down-regulating the expression of anti-apoptotic proteins bcl-2,caspase3,and caspase9destroys or even kills HepG2 cells,thereby promoting the apoptosis of HepG2 cells.(5)Research on the effect of rose total flavonoids on intestinal flora.RFW and RFE were applied to eight common intestinal bacteria,and the influence of rose flavonoids on the intestinal flora was judged by measuring the growth curve of probiotics,acid production capacity and inhibition rate of pathogenic bacteria.The results showed that RFW has a better effect on promoting the growth and reproduction of Lactobacillus paracasei,Lactobacillus acidophilus,Bifidobacterium lactis and Bifidobacterium bifidum,and can speed up the time to reach the end of culture.RFE has a better growth-promoting effect on Lactobacillus bulgaricus and Bifidobacterium bifidum.The inhibitory effect of RFW and RFE on Staphylococcus aureus is greater than that of Escherichia coli,and the effect of RFE is better than that of RFW.(6)Research on the application of rose total flavonoids to food preservation.Using potassium sorbate(PS)as a positive control,RFW and RFE were applied to fresh soy milk and fresh orange juice.The freshness preservation effects of RFW and RFE were judged by indicators such as the total number of colonies,p H value and sensory evaluation.The results showed that RFW and RFE had certain antibacterial properties,which can effectively inhibit the growth of microorganisms in food and prevent rancidity.Because RFE are slightly soluble in water solvents,it is easy to precipitate in beverages,which also affects the stability of its state.RFW can not only effectively prevent the growth of microorganisms but also increase the aroma of the drink while maintaining its stability.
