Fractional Extraction Technology Is Applied To The Precise Separation Of Active Ingredients In Chinese Herbal Medicine

Abstract/Summary:
The active components are the material basis of the physiological function of Chinese herbal medicine.The separation of active monomers with high purities from Chinese herbal medicine is of great significance to the development of new drugs and the elucidation of the therapeutic mechanism.Traditional separation methods cannot meet people’s requirements for high purity and high yield at the same time.Therefore,it is necessary to develop an efficient separation method to obtain pure active ingredients.In this work,the separation of chlorogenic acid from Eucommia ulmoides extract,daidzin and puerarin from Pueraria lobata root extract,paeoniflorin and paeoniflorin from peony has been studied.In liquid-liquid extraction,the effects of organic solvents,p H and other factors on the distribution coefficient(k)and separation factor(α)were investigated,and efficient extraction systems were constructed.Based on phase equilibrium and the law of material conservation,a mathematical model was established.The effects of the number of mass transfer units(NTU),volume flow ratio and feed position on the extraction performance were simulated.The residual rate was introduced to optimize the extraction process.1.The study of Chlorogenic acid was separated from Eucommia ulmoides.When isoamyl alcohol acted as the extraction solvent,k CAof 1.88 and k6of 0.65were obtained,and then the separation factor between them was 2.9.When NTU was10,and intermediate feeding was adopted,chlorogenic acid with purity of 97.2%and yield of 81.4%was obtained.The experimental and simulated values were in good agreement.When NTU was increased to 30,the yield of 99.26%would be obtained,and the purity was more than 98%.2.The study of daidzin and puerarin were separated from Pueraria crude extract.n-octyl alcohol-water was used as the extraction system for separation of daidzin from Pueraria crude extract,kdaidzinof 2.2 and kpuerarinof 0.6 were obtained,and the separation factor between them was 3.66.A 10-stage centrifuge was used to separate daidzin,daidzin with 98%purity was obtained,the yield is 71.6%.The experimental and simulated values were in good agreement.When NTU was 20,the yield and purity of daidzin were higher than 98%.A two-step method was used to separate puerarin from Pueraria crude extract.n-octanol-water was used as the extraction system I.When NTU was 20,the daidzin were basically removed,and the retention rate of puerarin was as high as 98%.n-butanol-water was used as the extraction system II.kpuerarinof 5 and kmonomer 6of 3 were obtained,and the separation effect was1.66.When NTU was 50,the purity was 98%and the yield was 98%.3.The study of paeoniflorin and albiflorin;were separated from crude peony extract.The mixed solvents of n-butanol and ethyl acetate was used as extraction systems for separating paeoniflorin and paeoniflorin from crude peony extract.n-butanol/ethyl acetate(1:4)-water as the extraction system I.When NTU was 20,impurity monomers with larger separation coefficient were removed,and the retention rate of paeoniflorin and paeoniflorin could be more than 98%.n-butanol/ethyl acetate(3:2)-water was used as extraction system II,when NTU was50,the yield of paeoniflorin and albiflorin with 99.5%would be obtained in the water phase and the organic phase,respectively.
