| The cynomolgus monkeys are increasingly attractive for use as a non-human primate animal model in biomedical research.Using transgenic primates to construct animal models of human diseases caused by single or multiple gene mutations can be useful to study the pathogenesis and treatment of human diseases.The establishment of a single or multiple gene mutation-induced cynomolgus monkey disease model relies on assisted reproductive technology and embryonic genetic modification.Studying the reproductive physiology of cynomolgus monkeys is valuable to improve the superovulation response efficiency of egg donor cynomolgus monkeys and explore the selection criteria of embryo transfer recipients.Therefore,we designed the current study to determine physiological indicators for effective screening of egg donors and surrogate cynomolgus monkeys.Experiment one:in this experiment,we randomly selected 12 adult female cynomolgus monkeys and detail about blood extraction,sexual skin observation,vaginal smear,colposcopy,B-ultrasound monitoring of the ovaries and uterus,laparoscopic observation of the ovaries,collected every seven days,with a continuous collection of about two menstrual cycles were collected and data was statistically analyzed.The results showed that sexual skin morphology,vaginal keratinocytes,vaginal mucosa and morphology were changed with follicular growth.In ovulation,there was E2 peaked(325.27±177.55 pg/m L)and LH peaked(25.88±17.69 ng/m L).Sexual skin has the largest degree of swelling and the darkest color in ovulation.The proportion of vaginal keratinocytes also peaks at the ovulation and is as high as 96.77%.Additionally,the vaginal mucosa has the highest degree of thickness during ovulation.These indicators can be used as a predictor of ovulation,providing an important basis for surrogate mother selection.Experiment two:in this study,we randomly selected 18 adult female cynomolgus monkeys,and were divided into two groups(I)superovulation good responders and(II)superovulation poor responders groups according to follicular development.Biological details were collected and data was statistically analyzed.The result showed that the LH level in the poor responders group on the first day of superovulation was significantly higher than that in the good responders group.The LH in the poor responders group on the sixth day of superovulation was still significantly higher compared to the good responders group,and the E2 level in the good responders group was significantly higher than that in the poor responders group.The sexual skin changes in the good responders group during the superovulation process were more obvious compared to the poor responders group.In the poor responders group,the B-ultrasound showed less clear follicles on the sixth day of superovulation,while the good responders group had clear and more dominant follicles.The other indicators did not change significantly among both groups.In summary,in this study we found judge ovulation by sexual skin and vaginal mucosa observation and vaginal smear beside according to the level of E2 and LH.We suggest that try to use individuals with obvious changes in the sexual skin during normal menstrual cycle and normal hormone levels on the first day of superovulation as egg donors.On the sixth day of superovulation,the ovarian response can be predicted according to the E2 level,or according to the degree of sexual skin changes and B-ultrasound observation of ovarian and follicular changes. |
