| In order to reduce the fluff limit of the Bothriochloa ischaemum seeds and ensure seed vigor,thereby speeding up its mechanized production and utilization efficiency,this experiment used Bothriochloa ischaemum seeds as materials,depilated the seeds with three chemical reagents(HCl、H2SO4 and NaOH),and four kinds of common reagents(MT、GSH、As A and PEG)were used to repair the seeds after velvet removal.Through the determination of seed germination percentage(Gp),germination index(Gi),average germination time(MGT),seedling vitality index(SVI),the determination of antioxidant enzymes and other related indicators after depilation and restoration,to study the effect of different depilation and repairing methods on seed germination and antioxidant capacity of Bothriochloa ischaemum,and to select the best defleecing and repairing methods of Bothriochloa ischaemum seeds.The main results obtained are as follows:(1)The seeds germination of Bothriochloa ischaemum was inhibited after HCl develveting,and the seed vigor level was generally low: Gp,Gi and SVI of seeds after develveting gradually decreased with the increase of HCl concentration and treatment time,and antioxidant enzyme activity generally increased first and then decreased with the increase of treatment time.(2)After H2SO4 develveting,the Gp,Gi and SVI of Bothriochloa ischaemum seeds decreased with the increase of the treatment time,and showed a trend of increasing first and then decreasing with the increase of H2SO4 concentration.After demining 60% H2SO4 for 1 min,the Gp,Gi and SVI of seeds were significantly(P<0.05)higher than the seeds not fluffed(CK1)and the germination ability was strong.However,the H2O2 contents in the seeds were significantly(P<0.05)higher than CK1,and the activities of SOD,CAT and APX(P<0.05)were significantly lower than CK1,and the antioxidant capacity was weak.(3)The Gp,Gi and SVI of Bothriochloa ischaemum seeds after NaOH develveting increased with the increase of solution concentration,but the activities of various antioxidant enzymes decreased with the increase of treatment time.(4)When repaired with MT with a concentration of 0.025 mmol/L for 12 h,the Gp,Gi and SVI of the seeds reached the maximum value of MT repair,Gp was 25% higher than the unrepaired velvet seed(CK2),and the H2O2 content in the seeds was significantly(P<0.05)lower CK2,POD and SOD activities were significantly(P<0.05)higher than CK2,and the seed vigor level was the highest.(5)When GSH with a concentration of 0.1 mmol/L was used for 20 h,Gp,Gi and SVI reached the maximum value of GSH repair,Gp was 12.52% higher than CK2,and the germination ability of the remaining GSH treated seeds was weaker than CK2.(6)When the concentration of As A was 2.0 mmol/L for 12 h,the maximum Gp,Gi and SVI of the seeds reached the maximum of As A repair,Gp was 19.25% higher than CK2,and the MDA content in the seeds was significantly(P<0.05)higher than other treatment times.(7)The seed germination ability of PEG repair is much weaker than CK2.The activities of CAT and APX in seeds without PEG repair were significantly(P<0.05)higher than those after repair.Therefore,PEG has the worst repairing effect on Bothriochloa ischaemum seeds after develing.In summary,the effects of three chemical reagents on develveting Bothriochloa ischaemum seeds are H2SO4>NaOH>HCl.The effects of the four reagents on the viability restoration of develvet seeds are MT>As A>GSH>PEG.Depilation treatment with 60% H2SO4 for 1 min followed by 0.025 mmol/L MT restoration treatment for 12 h is the best method to reduce fluff and improve seed vigor of Bothriochloa ischaemum in production practice. |
