| Plukenetia volubilis is a perennial woody oilseed vine,which belongs to Euphorbiaceae family,its seed oil content is 31-40%,rich in polyunsaturated fatty acids,it is one of the ideal raw materials for health edible oil.But currently the results of large-scale planting in various regions show that P.volubilis has weak resistance to disease,serious disease and large-scale production reduction,which affects its industrialization development.In addition,P.volubilis belongs to facultative cross breeding system,and the progeny of seed will produce certain phenotypic separation,through the agronomic traits evaluation and analysis of single plant of P.volubilis seed seedlings,excellent plant can be selected.In addition,In order to improve the genetic diversity of P.volubilis in China and speed up the cultivation of new varieties with disease resistance and high yield,based on the transcriptome data of P.volubilis,SSR markers were further developed.The main results are as follows1.Compared with the open-field cultivation,the flowering and fruiting times of P.volubilis were about 20 days later in the rain-shelter cultivation mode(high shed and low shed).The mortality of P.volubilis was low in the high shed cultivation mode which was 31.11 ± 0.17%,and the mortality rate was only 10.45 ± 0.06%in the low shed cultivation mode.The seed yield per plant was relatively high in the high shed cultivation which was 490.72±42.82 g,and it significantly decreased in the low shed cultivation which was 397.15 ± 19.47 g.There was no significant difference in single seed weight and oil content of seeds under different cultivation models in different harvest seasons2.Based on the analysis of the agronomic traits in P.volubilis from three geographical provenances,it was found that the high seed yield and single seed weight per plant was 1206.6 g and 1.44 g.The plants with high oil content of seeds is from Peru which was 41.62%.In addition,the plants with high seed yield and oil content were selected as candidate materials,of which seed yield per plant was higher than 817 g,single seed weight was more than 1.30 g and oil content was more than 40.00%.In order to produce high-yield and high-oil plants,we can further develop breeding and inter plant hybridization3.A total of 57664 unigenes were obtained from the transcriptome of male and female inflorescence buds in P.volubilis using MIS A software,and a total of 10572 SSR loci were detected.Among them,8825 unigenes have SSR loci,accounting for15.30%of the unigenes.The average distribution frequency of SSR loci was one SSR loci per 5.34 kb.The dominant motifs in the 154 SSR types were mononucleotide repeat,trinucleotide repeat and dinucleotide repeat,accounting for 40.48%,28.89%and 25.82%,respectively.A/T,AG/TC,and AAG/CTT were the dominant repeat motifs in mononucleotide,dinucleotide and trinucleotide types,respectively.Among the designed 75 pairs of SSR primers,31 primers could be amplified clear bands effectively,and 7 primers with polymorphism among P.volubilis,P.corniculata and P.conophora were finally obtainedThrough the evaluation of the agronomic characters of P.volubilis in the rain-shelter cultivation,the best planting pattern of P.volubilis in Xishuangbanna was selected;through the analysis of the agronomic traits of three geographical provenance strains of P.volubilis,the excellent provenance was provided for the future breeding of P.volubilis;in addition,based on the transcriptome data of P.volubilis,the SSR marker of P.volubilis was developed to identify the germplasm resources of P.volubilis molecular marker assisted breeding laid the foundation. |
