| To investigate the effects of traditional Chinese medicine(TCM)on anti-oxidative status and the expression of inflammatory cytokines in the piglets,eighteen crossbred(Durox×Landrace×Yorkshire)piglets(weight 21.43±2.86 kg)were randomly divided into three groups(Control,TCMⅠand TCMⅡ),and each group had 6 piglets.Piglets were fed with the basal diet,basal diet affixed with 10 g/kg Xiao Jian Zhong plus-minus compound(XJZ),and basal diet affixed with 3g/kg Jingsananli-sepsis powder(JJS)in the three groups,respectively.The serum was gathered on days 0,30 and 60 to determine the levels of biochemical indexes(TP,ALB,ALT,AST and ALP),antioxidant indicators(T-AOC,SOD,GSH-P_X and MDA)and inflammatory factors(TNF-α,IL-6,IL-8 and IL-10).Additionally,the liver samples were also collected on day 60 to detect the m RNA(SOD-1,HO-1,NQO-1,Nrf-2,NF-κB,IL-6,IL-8 and TNF-α)expression and protein(HO-1,Nrf-2,NF-κB,IKB-α,p-NF-κB and p-IKB-α)expression.The experiment result is given as:1.Compared to the Control group,the activity of ALT,AST and ALP were significantly decreased on day 60 in the TCMⅠgroup(P<0.05 or P<0.01),while the concentrations of TP and ALB have no difference in the TCMⅠand TCMⅡgroups on days 30 and 60(P>0.05).2.Compared to the Control group,the content of T-AOC was significantly increased on days 30 and 60 in the TCMⅠand TCMⅡgroups(P<0.05 or P<0.001),the level of GSH-P_Xwas significantly increased on day 60 in the TCMⅠgroup,while the MDA concentration was significantly decreased in the TCMⅠand TCMⅡgroups on day 60(P<0.05).3.Compared to the Control group,the levels of TNF-αand IL-6 were significantly decreased in the TCMⅠgroup on day 60(P<0.05),IL-8 concentration was significantly decreased in the TCMⅠand TCMⅡgroups(P<0.05 or P<0.001).4.Compared to the Control group,the expression of Nrf2,SOD-1,NQO-1,and HO-1m RNAs were significantly increased(P<0.05 or P<0.001),while the expression of NF-κB,TNF-α,IL-6,and IL-8 m RNAs were decreased in TCMⅠand TCMⅡgroups(P<0.05 or P<0.001).5.Compared to the Control group,the protein levels of Nrf2 and HO-1 were significantly increased(P<0.001),while the phosphorylation levels of NF-κB and IκB-αwere significantly decreased in TCMⅠand TCMⅡgroups(P<0.001).Collectively,these findings suggested that TCMⅠand TCMⅡup-regulated anti-oxidative and anti-inflammatory capabilities in the nursery piglets,which might be modulated by the Nrf2/NF-κB signaling pathways. |
