At present,the production of domestic dog anesthetic preparations is subject to certain restrictions.The available varieties are single,have large side effects,and have a high incidence of clinical accidents.It is difficult to meet the needs of clinical diagnosis and treatment.Compound anesthesia is a common anesthesia method in clinical practice.Several drugs are used together to meet the anesthesia needs,which can not only effectively reduce the dosage of each drug,but also avoid many side effects.In this trial,thiprazole,teltamine,and midazolam were combined into a mixture to study the anesthetic effect of this compound anesthesia on dogs and the safety of application in dog anesthesia.Through the combination of different drugs in different proportions,preliminary screening and scientific prescription experiments were conducted to screen the approximate proportions of thiprazole,teltamine and midazolam as composite anesthetic preparations;by conducting three factors on mice Three-level orthogonal test,anesthesia effect evaluation and safety evaluation test(LD50 and ED50)applied to dogs,the best dosage of compound anesthesia preparation was screened out and applied to the dog’s routine physiological surgery to evaluate the anesthesia effect.Orthogonal test and canine anesthesia monitoring and evaluation results show that thiazolazole: teltamine: midazolam 1.2: 0.3: 0.5(calculated based on the volume of a single injection in the mixture)In the orthogonal experiment,the maximum effective rate of righting reflection is 100%,In the trial optimization combination,the shortest anesthesia time was 46 min,the longest anesthesia time was 159 min,It has good sedation,analgesia and muscle Relaxation effect;the safety test results of mice show that the safety factor is 11.5,indicating that the composite anesthetic has a large safety range and can meet the basic needs of animal clinical anesthesia.The compound anesthesia was used for the operation of dogs.The results showed that the mixture can provide anesthesia time of about 120 minutes,which can meet the clinical routine surgical operations,such as laparotomy,castration and sterilization.Compared with the operation group,In the non-surgical group,the decrease in body temperature and blood pressure was more significant,and there was no significant difference between the two groups at the same time point.During the operation,the dog’s heart rate(HR),respiratory rate(RR),and blood oxygen saturation(SPO2)all showed a trend of decreasing first and then slowly increasing.Based on the above results,it can be concluded that after the thiazolazole compound preparation is applied to dogs,the anesthesia has a fast onset,anesthesia is maintained for a long time,an appropriate depth of anesthesia(no struggling during anesthesia),and can provide better analgesia and sedation And muscle relaxation effect,anesthesia effect is indeed.Thiazolazole compound preparation has little effect on various physiological indexes during the physiological anesthesia of dogs,can meet the needs of clinical anesthesia,and can be used as a potential compound anesthetic agent for dogs.
