Tremella aurantialba,belonging to Tremellales,Tremellaceae,Tremella in The Classification Status,is a kind of rare edible fungi with best development potential.In recent years,it has been loved by people.The market demand of Tremella aurantialba is increasing year by year,and the supply is in short supply.The key step for producing Tremella aurantialba is preparation of spawn.At present,the majority of producers use solid spawn,directly inoculating the mixed culture of Tremella aurantialba and Stereum hirsutum.The ratio of the two strains is difficult to be controlled artificially,and the quality of strains is uneven,leading to the phenomenon that no mushrooms can be produced after cultivation,and the yield of fruiting body is not stable.In addition,the current production mode of Tremella aurantialba is mainly cultivated with basswood,which has a lot of disadvantages,such as high cost and harmful to the environment,and the problems of group research were all affecting the sustainable development of Tremella aurantialba industry.Therefore,it is of great significance to study the culture of liquid spawn and Tremella aurantialba cultivated with substitute for the liquid spawn producing and its large scale cultivating.This study separated,purified and identified the fruiting body of Tremella aurantialba.Through the cultivation method of shake flask,the liquid medium formula and culture conditions of Tremella aurantiabla and Stereum hirsutum were optimized,which determined their optimum conditions.And the correlation between the inoculation method,mixing ratio,mixing time and the growth and development of Tremella aurantialba was carried out.In addition,the culture substrate and culture conditions of Tremella aurantialba were studied so as to explore the best culture substrate formula and culture conditions.The main research results are as follows:1.Strain J-1 was identified as Tremella aurantiabla,and strain M-1 was identified as Stereum hirsutum by combining with morphological characteristics and molecular biological identification.2.The optimum medium formula and culture conditions of Stereum hirsutum were determined through single factor experiment,Plackett Burman design experiment,the steepest ascent test,and response surface methodology:yeast extract powder 6.7 g/L,soluble starch36.6 g/L,K2HPO42.4 g/L,KH2PO42.0 g/L,VB120.0 mg/L.The initial p H was 6.0,the the culture temperature was 24℃,and the volume of liquid was 100 m L.The optium liquid culture medium formula and culture conditions of Tremella aurantialba were as follows:sucrose 36.4 g/L,peptone 2.0 g/L,KH2PO44.8 g/L,KCl 2.9 g/L,Mg SO4·7H2O 2.0 g/L,VB610.0 mg/L.The initial p H was 6.7,the culture temperature was 24℃,inoculum size was 2%,and the volume of liquid was 80 m L.3.The results showed that the best inoculation method was to inoculate liquid spawn of Tremella aurantialba and liquid spawn of Stereum hirsutum simultaneously.When the mixing ratio of liquid spawn of Tremella aurantialba and liquid spawn of Stereum hirsutum was 1:1(v/v)and the mixing time was 1 day,the best experimental effect was obtained.Under these conditions,the growth rate of mycelium was 0.492 cm/d,the mycelium of Tremella aurantialba was white and grew relatively well,the time of mycelium growing all over the bag and primordium formation were 24 days and 17 days,respectively.The fruiting rate of Tremella aurantialba was 100%,the fruiting body yield and biological efficiency were 129.7g/bag and 64.85%,respectively.4.As a result,it showed that formula F was the best culture medium formula.The formula was as follows:cottonseed hull 78%,wheat bran 18%,corn flour 2%,gypsum powder 1%,Mg SO4·7H2O 0.5%,KH2PO40.5%.The optimal cultivation conditions for the growth of Tremella aurantialba mycelium were water content 60%,p H 7,liquid spawn inoculum size 15 m L.The optimum culture temperature for the formation of fruit body of Tremella aurantialba is 24℃.Through this study,the cultivation of Tremella aurantialba using liquid spawn was realized,which provided important reference value for the production of Tremella aurantialba,and promoting the scientific development of Tremella aurantialba industry.
