The cultivation of Agaricus bisporus by crop straw is one of the most important ways of straw resource utilizations.Compared with solid spawn,liquid spawn can effectively reduce the breeding time and improve the productions efficiency.The cultivation of A.bisporus by traditional fermentation materials,the composting fermentation processes are cumbersome,the time is long,and the fermentation quality is not easy to control,but the clinker cultivation is not subjected to seasonal restrictions,low pollution rates,stable productions,and easy to operate.In this paper,the interactions of each factor between the cultivation of Liao A strain,in liquid were optimized by RSM,to determine the optimum culture conditions,the physicochemical properties of fermentations were studied in different periods,and the cultivation of A.bisporus was carried out with liquid spawn.The main results obtained are as follows:(1)According to the physiological characteristics,the analysis of the sequence of ITS and the similarity of GenBank sequence of Liao A strain,the strain Liao A was identified as a kind of A.bisporus.Through the Bavendamm color reaction,Aniline blue decolonization reaction and Ander and Eriksson color ring test,it was clear that Liao A strain had certain lignin degradation ability.(2)On the bases of single factor,the culture conditions.of Liao A liquid were optimized by Plackett-Burman design and RSM.After the main influencing factors integer,the optimized culture conditions were(/L):corn flour concentration 15g,soybean meal concentration 10g,VB1 concentration 20mg/L,KH2PO4 concentration 3g,MgSO4 concentration 2g,initial pH 7.0,inoculation dose 5%,volume 77mL in 250mL triangular flask,culture temperature 26 ? shaker speed 120 r/min,and the culture cycle of liquid spawn was 7 days.At this time,the maximum dry weight of mycelium was 1.112g/100mL,which was 12.72%higher than before optimization.(3)The contents of culturable microorganisms in different periods of composting were determined by the dilution plate method.It was found that the number of culturable microorganisms had a change trend of increasing first and then decreasing.Onion ketone,coomassie brilliant blue G250 and other methods were used to analyze the soluble sugar contents,soluble protein contents,pH value and moisture contents.The results showed that soluble sugar contents of materials found in the composting progresses first increased and then decreased,soluble protein contents and pH value were increased,while the moisture contents decreased.(4)Six formulations were designed for clinker cultivation:100%straw,5%fermentation+95%straw,10%fermentation+90%straw,15%fermentation+85%straw,20%fermentation+80%straw,25%fermentation+75%straw,with the clinker fermentation materials as control,inoculating 8%Liao A liquid spawn for clinker cultivation tests,it was found that the 20%fermentation+80%straw clinker cultivation material,fruiting best,and the yield was about 90.2%of the clinker fermentation material.However,the yield of 100%straw clinker cultivation material is not ideal,to be further research.
