| With the development of the national work of reducing and replacing antibiotics,the development of green and healthy feed additives that can replace antibiotics has become the most important task.Many studies have found that natural green plants,especially food and drug homologous plants,have significant effects on promoting the healthy growth of animals and improving the antioxidant capacity of the body.Many factors in animal production can cause cell lipid peroxidation,lead to biofilm damage,hinder normal physiological and biochemical functions,and thus endanger animal health.Therefore,it is feasible to add natural plant ingredients in animal feed to improve the antioxidant capacity of animal lipid.After preliminary screening in the laboratory,this study selected the extract of Lycium barbarum and Astragalus membranaceus as feed additive,and took Sichuan Tibetan black pig as the experimental object to study the effect of the extract of Lycium barbarum and Astragalus membranaceus on the lipid antioxidant capacity of Sichuan Tibetan black pig.The lipid content and antioxidant index of longissimus dorsi of Sichuan Tibetan black pig were explored The relationship between meat performance and meat quality,growth performance.So,in this experiment,the 50 healthy Sichuan Tibet black pigs with 175 days old and weighed(70 ± 2)kg were randomly divided into two groups,with 5 replicates in each group and 5 pigs in each replicate.The control group was fed the basal diet,and the experimental group was fed the basal diet supplemented with 0.1% Lycium barbarum-Astragalus membranaceus Extract.The feeding period was 90 days.The longissimus dorsi muscle was taken between the last third and the fourth ribs on the left side of the carcass to detect pork quality,antioxidant index and transcriptome analysis.The results show that:(1)Growth performance: the slaughter weight,average daily gain and average daily feed intake of the experimental group were significantly increased(P < 0.05),which were 1.06 times,1.19 times and1.13 times higher than those of the control group.(2)Pork quality: compared with the control group,the scores of meat color and marbling in the experimental group were significantly increased by 19% and 52% respectively(P < 0.05);the contents of vitamin E and crude fat in the experimental group were significantly increased by 2.3 times and 1.1times respectively(P < 0.05);the total amount of unsaturated fatty acids in the experimental group was significantly higher than that in the control group by 1.9 times(P < 0.05).(3)Lipid antioxidant index: CAT activity and GPX activity of the experimental group were significantly increased by 1.1 times and 1.3 times(P < 0.05);GSH content of the experimental group was significantly increased by 1.5 times(P < 0.05),GSSG content was significantly decreased by 1.1times(P < 0.05),GSH / GSSG ratio was significantly increased by 1.6 times(P < 0.05).Compared with the control group,the content of LPO and MDA in the experimental group were significantly reduced by 1.4 times and 1.1 times(P < 0.05).(4)High throughput transcriptome data analysis showed that GCLC,GSS,GPx4,cat,LOX and other genes related to oxidative stress had significant changes.Go and KEGG analysis showed that they were mainly concentrated in the glutathione metabolic signaling pathway.From ELISA test,the content of GCLC,GSS and GPx4 in the experimental group was significantly higher than that in the control group by 12%,8% and 16%(P < 0.05).Conclusion: adding 0.1% Lycium barbarum-Astragalus membranaceus Extract to basic diet can improve glutathione metabolism and lipid antioxidant capacity,so as to promote healthy growth and meat quality of Sichuan Tibet black pigs. |
