OBJECTIVE:We detected the effect of the contents of oxidized lo w-density lipoprotein(ox-LDL)in rat serum and the expression of nucle ar factor-κB(NF-κB)in abdominal aorta tissue,in order to investigate th e protective functions of Sappanwood ethyl acetate extract suspension o n vascular endothelial cells.METHODS:40 male Sprague-Dawley rats were divided into blank c ontrol group,model control group,sappan group and simvastatin group by random method.The blank control group was fed with normal feed,and t he other groups were fed with 3%methionine feed.After 12 weeks,the s appanwood ethyl acetate extrac group was given intragastric of sappanw ood ethyl acetate extract suspension,and the simvastatin group was give n intragastric of simvastatin suspension,while the blank control group a nd the model control group was given intragastric of the same volumeo f sodium carboxymethyl cellulose suspension,once a day continuous 4 w eeks.Observed the changes oft he general condition of the rats,and the p athological changes of the abdominal aortic endothelium morphology by HE staining.The serum ox-LDL was detected by ELISA and the expressi on of NF-κB in the rat abdominal artery tissue was observed by immun ohistochemistry.RESUILTS:1.Pathological morphological changes in rat abdominal artery:The th ree-layer membrane structure of vascular tissue in the sappanwood ethyl acetate and simvastatin groups was complete.The endothelial cells were slightly swollen,the surface was less smooth,and the arrangement of smo oth muscle cells was slightly disordered.Compared with the model contr ol group,the pathological damage was alleviated.2.Rat serum ox-LDL results:Compared with the blank control group,the serum ox-LDL levels in the model control group were significantly higher(P<0.05);Compared with the model control,serum ox-LDL levels in the sappanwood ethyl acetate extract group and the simvastatin groupwe re significantly lower(P<0.05);Compared with simvastatin group,the seru m ox-LDL of sappanwood ethyl acetate extract group was higher,but the difference was not statistically significant(P>0.05).3.Expression of NF-κB in rat abdominal artery:Compared with the blank control group,the expression index of NF-κB in abdominal arteryo f the model control group,the sappanwood ethyl acetate group,and the si mvastatin group were increased(P<0.05);Compared with the model contr ol,the expression index of NF-κB in abdominal artery of the sappanwoo d ethyl acetate extract group and the simvastatin group were significant ly lower(P<0.05);but compared with the simvastatin group,the expression index of NF-κB in abdominal artery of sappanwood ethyl acetate extract group was lower,the difference was not statistically significant(P>0.05).CONCLUSIONS:1.The sappanwood ethyl acetate extract could reduce the damage o f endothelial in endothelial injury model of rats.2.The sappanwood ethyl acetate extract could reduce the levels of serum ox-LDL in rat models of vascular endothelial injury.3.The sappanwood ethyl acetate extract could reduce the expression of NF-κB in abdominal artery.
