In order to make traditional Chinese medicine globalization and modernization,it is necessary to improve the quality control of traditional Chinese medicine.The quality of traditional Chinese medicine will directly affect the clinical efficacy of its products,how to improve the quality and stability of traditional Chinese medicine,has been subject to the industry personnel’s high attention.The 2015 edition of the Chinese Pharmacopoeia(Part Ⅰ)provides that Chuanxiong Rhizoma comes from the dried rhizomes of Ligusticum chuanxiong Hort.,have the effect of invigorate the circulation of qi and the functions of the pain,―upward leader,descending blood‖,as the famous Sichuan common commodities production of medicinal materials,is a commonly used drug for the treatment of disease of heart head blood-vessel.Traditional Chinese medicine theory has always emphasized the sensory evaluation of ―large size,solid quality,yellow and white section,large oil and rich aroma‖ of Chuanxiong Rhizoma,and the clinical and market circulation of Chuanxiong Rhizoma drugs of different grades of mixed use and mixed sale of size,has a enormous impact on the clinical use of Chuanxiong Rhizoma safety.At present,ferulic acid is only used as the index in the quality control of Chuanxiong Rhizoma in the 2015 edition of Chinese pharmacopoeia.Ferulic acid also exists in other medicinal materials such as Angelica sinensis(Oliv.)Diels.,and its specificity is not strong for Chuanxiong Rhizoma,which makes it difficult to reflect the comprehensive quality of it.In this study,different grades of Chuanxiong Rhizoma were taken as the research object.Based on the traditional Chinese medicine theory,sensory analysis and characteristic component analysis were carried out on the appearance of Chuanxiong Rhizoma,different grades of Chuanxiong Rhizoma were identified by machine learning.The color and odor were correlated with the characteristic components,providing a new method for the quality evaluation of Chuanxiong Rhizoma.Objectives1.Sensory identification of odor and color characters of Chuanxiong Rhizoma was conducted based on the traditional Chinese medicine theory.2.Distinguish different grades of Chuanxiong Rhizoma,evaluate the quality difference between its color odor and characteristic components,and analyze the correlation between color odor and characteristic components of different grades of Chuanxiong Rhizoma.Methods1.Digital analysis and identification of ―smell‖ and ―color‖ of Chuanxiong Rhizoma were carried out by using electronic nose and machine vision color analysis technology.RGB color space was selected to describe the color of Chuanxiong Rhizoma of different grades.2.The volatile components of Chuanxiong Rhizoma of different grades were analyzed by HS-GC-MS,and the odor material basis of it was analyzed by electronic nose technology.3.The methods of near infrared spectroscopy and stoichiometry were used to distinguish different grades of Chuanxiong Rhizoma.4.Using HPLC method,to 85 batches of Chuanxiong Rhizoma of different grades of ferulic acid,levistolide A,senkyunolide A,and Z-ligustilide content of four components.5.The contents of ferulic acid,levistolide A,senkyunolide A,and Z-ligustilide in different grades of L.chuanxiong.were detected by HPLC,respectively,as well as the maximum response value of electronic nose odor and RGB value of color,and correlation analysis was conducted by statistical method.Results1.The odor fingerprint map of Chuanxiong Rhizoma based on PEN3 electronic nose was established.The odor response value characteristic map of different grades of chuanxiong rhizome’s electronic nose was different,and W1 W,W2W,W5 S and W1S4 sensors in PEN3 sensors were sensitive to Chuanxiong Rhizoma’s odor.Using machine vision color analysis technology,the method of RGB color space analysis of Chuanxiong Rhizoma was established.2.The volatile compounds of ligusticum chuanxiong were analyzed by HS-GC-MS,and a total of 71 compounds were identified,among which 26 compounds were related to different grades of Chuanxiong Rhizoma.The compound with the highest relative content was Z-ligustilide.The results show that the volatile compounds in different grades of Chuanxiong Rhizoma are different.HS-GC-MS and electronic nose analysis were used to identify the important compounds with different grades of Chuanxiong Rhizoma: 5-pentylcyclohexa-1,3-diene;alpha-Cedrene;1HCycloprop[e]azulene,1a,2,3,5,6,7,7a,7b-octahydro-1,1,4,7-tetramethyl-,(1a R,7R,7a S,7b R)-;2-metoxy-4-vinyl-phenol.3.Compared with SVM algorithm,ELM can better distinguish Chuanxiong Rhizoma quality grades,more than 80 % correct,including the HS-GC-MS to detect the contents of the volatile compounds of relative test set up to an accuracy of 89.95 %.The result of the fusion of electronic nose and machine vision data was better than that of single odor and color data.The accuracy of the test set was 89.00 %,indicating that the odor and color of Chuanxiong Rhizoma and the volatile compounds could best distinguish the different grades.4.To establish A HPLC method for simultaneous determination of ferulic acid,levistolide A,senkyunolide A,and Z-ligustilide in Chuanxiong Rhizoma.The contents of different grades of Chuanxiong Rhizoma are different.5.Correlation analysis of the contents of chemical components with odor and color in different grades of Z-ligustilide and ferulic acid in different grades of Chuanxiong Rhizoma showed that there was a significant correlation between the contents of Z-ligustilide,ferulic acid in different grades of Chuanxiong Rhizoma and W2 S sensor in electronic nose(P<0.05),and there was a significant correlation between the contents of Z-ligustilide and ferulic acid in different grades of Chuanxiong Rhizoma and W2 W sensor in electronic nose(P<0.05).The content of ferulic acid was significantly correlated with the color R value and G value(P<0.05),and the content of Z-ligustilide was significantly correlated with the color R value(P<0.05).The contents of ferulic acid and levistolide A,and Z-ligustilide in different grades can be determined by means of electronic nose and color RGB value.Conclusions1.The objective quantification method of the odor and color of Chuanxiong Rhizoma by electronic sensory technology was established.The correlation analysis between the content of characteristic components,the color and the odor was analyzed,so as to provide an effective reference for the further study on the correlation between the appearance and quality of Chuanxiong Rhizoma.2.Sensory recognition of color and odor combined with characteristic component analysis,the identification model of different grades of Chuanxiong Rhizoma was established by machine learning,providing a new method for the integrated quality evaluation of Chuanxiong Rhizoma. |
