Protective Effects Of L-Glutamine On Acute Renal Ischemia-reperfusion Injury And High-throughput Sequencing Analysis Of MicroRNAs

Abstract/Summary:
Objective:To investigate the effect of L-Glutamine on acute renal ischemia-reperfusion injury and the differential expression of Mirna after L-Glutamine intervention by establishing rat model of acute renal ischemia-reperfusion injury and detecting the levels of blood urea nitrogen,creatinine and renal pathology,to predict the involvement of related target genes in the signaling pathway,and to provide a new idea for targeted therapy.Method:Rats were randomly divided into 5 groups.Nor+NS Group:NS was injected without laparotomy.Sham+NS and Sham+GLN Group: the right kidney was resected,the left renal pedicle was not clamped,the same amount of NS or GLN was injected.In IR+NS Group and IR+GL Group,the right kidney was resected,the renal artery was clamped,and the same amount of NS or GLN was injected after successful reperfusion.At 1h,5h,12 h and 24 h after modeling,renal tissues were stained with HE,Scr and BUN were detected,and Mirnas were differentially expressed by high throughput sequencing,and analyzed by KEGG and GO.Results: Compared with I/R+NS,I/R+GLN group had less pathological changes in renal tubules after 24 hours of I/R+NS,creatinine and urea nitrogen levels in I/R+NS Group increased continuously from 1 hour to 24 hours,and reached the peak at 24 hours,compared with Nor+NS Group and Sham+NS Group,the difference was statistically significant(p<0.05).The urea nitrogen level of I/R+GLN Group was significantly higher than that of Sham+GLN group at each time point(p<0.05).The Serum Creatinine of I/R+ GLN Group had no significant difference compared with Sham+GLN group at each time point.The serum creatinine and Urea Nitrogen in I/R+GLN group increased from 1 hour to 12 hours,but the creatinine and urea nitrogen in I/R+NS group increased slowly,and the creatinine and urea nitrogen levels at each time point were lower than those in I/R+NS Group(p<0.05).Hippo signaling is involved in both SHIN-vs-SHIG and SHIN-vs-SHSN.The L-Glutamine may be associated with Hippo signaling in the process of alleviating ischemia-reperfusion induced acute kidney injury.Conclusion: 1.L-Glutamine relieves acute renal injury caused by ischemia-reperfusion.2.There were significant differences in Mirnas expression after ischemia-reperfusion injury.3.L-glutamine attenuates ischemiareperfusion-induced acute kidney injury in a process that may be associated with the Hippo signaling pathway.
