ObjectiveTo explore the differences of heat shock protein 90 levels in the aqueous humor of patients with different types of cataracts, and analyze the correlation between the contents of heat shock protein 90 in the aqueous humor and the severity of cataract patients,so as to provide theoretical basis for early screening and targeted treatment of different types of cataract in the future.MethodsFrom April 2019 to December 2019,the patients who were diagnosed as cataract and treated by phacoemulsification combined with intraocular lens implantation in the First Affiliated Hospital of Zhengzhou University were selected, and the patients diagnosed with the premacular membrane without cataract were used as the negative control group.According to the causes of cataract, the cataract patients were divided into age-related cataract group, diabetic cataract group, high myopia cataract group and negative control group. The content of HSP90 in aqueous humor was determined by Western blot and ELISA.Spss22.0 statistical software was used to analyze the data, and the measurement data were expressed as mean±standard deviation.Descriptive statistical analysis and one-way ANOVA were used to analyze the content of HSP90 in aqueous humor of different experimental groups, such as age-related cataract group, diabetic cataract group, high myopia cataract group and negative control group The correlation of Hsp90 content in aqueous humor with visual acuity, intraocular pressure and glycosylated hemoglobin was analyzed. The risk factors of cataract were discussed by multivariate logistic regression analysis. The ROC curve was drawn by Graphpad Prism 8 software to analyze the predictive value of visual acuity,Hsp90 and slit lamp in different types of cataract groups. The difference was statistically significant(P<0.05).Results1.The levels of HSP90 in aqueous humor of age-related cataract group, diabetic cataract group and high myopia cataract group were 201.25±71.53(pg/ml),291.63±63.27(pg/ml)and 192.49 ±23.58(pg/ml)respectively,which were significantly higher than 60.15±8.36(pg/ml)of negative control group(P<0.05).Meanwhile,the expression of HSP90 in aqueous humor of diabetic cataract group was significantly higher than that of high myopia cataract group and age-related cataract group(P<0.05).However,there was no significant difference in the expression of HSP90 in aqueous humor between high myopia cataract group and age-related cataract group(P>0.05);3.There was a negative correlation between visual acuity and HSP90(r=0.524,P<0.05);there was no significant correlation between intraocular pressure and HSP90(P>0.05).4.Gender,age,visual acuity,HbAlc,intraocular pressure and HSP90 were included in the multiple logistic regression equation. The results showed that the final variables included in the equation were age,HbAlc and Hsp90,or were 3.976,3.456 and 5.197,P<0.05,The results showed that age,HbAlc and HSP90 increased by 3.976 times,3.456 times and 5.197 times when other variables remained unchanged.5.ROC curve was used to analyze the predictive value of visual acuity,Hsp90 and slit lamp in different types of cataract. The results showed that the AUC of visual acuity was 0.730,P<0.05;the AUC of Hsp90 was 0.804,P<0.05;the AUC of slit lamp was 0.926,P<0.05.ConclusionThe levels of heat shock protein 90 in aqueous humor of patients with different types of cataract are different, and they are higher in patients with diabetic cataract and traumatic cataract;age, expression of heat shock protein 90 and HbAlc are risk factors of cataract;visual acuity, refractive index, heat shock protein 90 and slit lamp can be used as predictors of cataract severity The order of predictive value from high to low was slit lamp detection>heat shock protein 90>visual acuity>refractive index.
