Abstract/Summary:
Objective:By observing the effects of transcutaneous electrical acupiont stimulation(TEAS)on bone mineral density,bone biomechanics,bone histomorphology and tumor necrosis factor-a(TNF-α)expression in rabbits with ovariectomized osteoporosis(PMOP),we explored a possible mechanism of TEAS in the treatment of PMOP.Methods:Thirty-six healthy female rabbits were randomly divided into three groups: TEAS group(n=8),model control group(n=14)and sham operation group(n=14).The PMOP model was replicated by bilateral oophorectomy in TEAS group and model control group,and the PMOP model was established by bilateral false oophorectomy in sham operation group.After 12 weeks of modeling,rabbits in the transcutaneous electrical acupiont stimulation group were treated with TEAS,while the other two groups were fed and managed together with the TEAS group without special intervention.At the same time record the weight of three groups of rabbit,mental state,activity,hair,etc.In general,in before and after 8 w treatment on bone mineral density,bone biomechanics,bone tissue morphology test,in the treatment of 4 w and 8 w after treatment using enzyme-linked immunosorbent method to detect the expression of TNF alpha level,and by using SPSS statistical software for data processing and analysis.Results:1.The building after completion of 12 w rabbit model control group(third density was 0.256±0.026(g/cm2),third(density control the rabbit was 0.336±0.035(g/cm2),compared with the control group,model control group rabbit bone > 2 sd,osteoporosis diagnosis standard,and compared with control group,model control group significantly lower bone density(P < 0.05),suggesting building success.After 8weeks of treatment,the BMD in TEAS group was significantly higher than that in model control group(P=0.012<0.05).2.Before modeling,the body weight of the sham operation group was2.291±0.134(kg),the body weight of the model control group was 2.310±0.135(kg),and the body weight of the TEAS group was 2.291±0.134(kg),and there was no difference between the three groups(P>0.05).Before treatment,the body weight of rabbits in the sham operation group was 3.228±0.113(kg),that in the model control group was 3.400±0.159(kg),and that in the TEAS group was 3.388±0.164(kg).Before treatment,compared with sham operation group,the body weight of rabbits in TEAS group and model control group increased significantly(P<0.05).There was no significant difference in body weight between model control group and TEAS group(P>0.05).After 8 weeks of treatment,the body weight of rabbits in the sham operation group was 4.089±0.141(kg),the body weight of rabbits in the model control group was 4.375±0.249(kg),and the body weight of rabbits in the TEAS group was 4.131±0.197(kg).Compared with the model control group,the body weight of rabbits in the sham operation group and the TEAS group increased slowly(P<0.05).3.During the adaptive feeding period,rabbits in the three groups had smooth hair color,normal mental and activity.After modeling,rabbits in each group were slow in activity and listless in spirit.During the treatment period,the rabbits in the model control group had sparse hair,slow activity and relatively slow response.The hair of the sham operation group and the TEAS group was smooth,and the mental activity was normal.4.Before treatment,the bone mechanics of the model control group was significantly different from that of the sham operation group.The maximum breaking force,rigid strength and maximum displacement before breaking were significantly reduced,compared with that of the sham operation group,P<0.05;After the intervention of TEAS,compared with the model control group,the maximum breaking force,rigid strength and the maximum displacement before breaking in the TEAS group were significantly increased(P<0.05).5.Before treatment,compared with the sham operation group,the trabecular density and the trabecular thickness of the model control group were significantly decreased(P<0.05);After TEAS intervention,compared with model control group,the bone trabecular density and bone trabecular thickness in TEAS group were significantly increased(P<0.05).6.After 4 weeks of treatment,TNF-α in model control group was significantly increased compared with sham operation group(P<0.05).Compared with model control group,TNF-α in TEAS group was decreased(P<0.05).After 8 weeks of treatment,TNF-α in TEAS group was significantly decreased compared with model control group(P<0.05).Conclusion:1.PMOP model could be successfully replicated by bilateral oophorectomy at12 W.2.TEAS can properly inhibit the abnormal weight gain caused by the imbalance of estrogen secretion.It can improve the mental state,activity and other general conditions of PMOP rabbits.By improving bone biomechanics,bone structure and bone density,it can achieve the effect of treating PMOP.3.TEAS can inhibit the expression of TNF-α in serum of PMOP rabbits.4.The treatment of PMOP by TEAS may be achieved by inhibiting the expression of TNF-α,reducing bone resorption and improving the bone metabolic environment to improve bone mineral density.However,the specific regulatory pathways need to be further studied.
