| Objective:Bi-min-Tong-qiao decotion(BMTQD)is a clinical treatment experience of allergic rhinitis(AR)in children,clinical trials confirm BMTQD effectively,this research intends to set up the AR model mice,detecting serum immunoglobulin E(immunoglobulin E,IgE)and eosinophil(eosinophil,EOS)and nasal mucosa tissue chemokine CCR3,CCL5,CCL11 expression,From the perspective of immune regulation,the effective effect and possible mechanism of BMTQD in regulating chronic inflammation of nasal mucosa in AR model mice were investigated.method:Using ovalbumin(OVA)sensitization and excitation,intranasal approach to establish BALB/c female mice model of allergic rhinitis were randomly divided into blank group,model group,BMTQD group:the high,medium and low dose and montelukast sodium group.Behavioral score of mice in each group;Hematoxylin-Esoinstaining(HE)method was used to evaluate the histopathology of nasal mucosa of mice in each group.The relative expression of IgE in peripheral blood of each group was determined by ELISA.Using flow cytometry technique(FCM)detection in peri pheral blood mononuclear cell(PBMC)eosinophil percentage;Immunohisto chemistry(IHC)was used to detect the protein expression levels of CCR3,CCL5 and CCL11 in nasal mucosa tissues of mice in each group.Results:(1)The behavioral scores of mice in model group were significantly higher than those in blank group(P<0.05);The behavioral scores of mice in montelukast sodium group and BMTQD groups were lower than those in model group(P<0.05);(2)Pathological scores of the model group were significantly higher than that of the blank group(P<0.05),and those of the montrukast sodium group and the BMTQD medium-dose group were significantly lower than that of the model group(P<0.05);EOS count in model group was significantly higher than that in blank group(P<0.05).Microscopy EOS count in montelukast sodium group and BMTQD groups were lower than that in model group(P<0.05);(3)The relative expression level of total IgE in serum of model group was significantly higher than that of blank group(P<0.05);The relative expression level of IgE in montelukast sodium group and BMTQD group were lower than that in model group(P<0.05);(4)The percentage of EOS in PBMC of model group was significantly higher than that of blank group(P<0.05);The percentage of EOS of PBMC in montelukast sodium group and BMTQD group were lower than that in model group(P<0.05);(5)The protein content of CCR3 in nasal mucosa of mice in model group was significantly higher than that in blank group;The content of CCR3 protein in nasal mucosa of mice in montelukast sodium group and BMTQD groups were significantly lower than that in model group(P>0.05);(6)The protein contents of CCL5 and CCL11 in nasal mucosa of mice in model group were significantly higher than those in blank group(P<0.05);CCL5 and CCL11 in nasal mucosa of mice in montelukast sodium group and BMTQD groups were lower than that in model group(P<0.05).Conclusion:(1)BMTQD can improve the inflammatory injury of nasal mucosa epithelial cells;(2)BMTQD have clear medicine efficacy which can down-regulate the high expression of IgE and EOS,alleviate the allergic reaction of AR model mice;(3)BMTQD can effectively improve the EOS infiltration of nasal mucosa in AR model mice and inhibit nasal mucosa inflammation,which is related to the expression of CCR3,CCL5 and CCL11. |
