Study On Anti-Inflammatory And Analgesic Effect Of Yao Medicine Jiu-Long Shi-Ba-Zheng Decoction,and Its Effect And Mechanism On Gastric Ulcer(Stomach Heat Syndrome)

Objective:To determine the safety and analgesic and anti-inflammatory effects of Jiu-long Shi-ba-zheng Decoction.In order to preliminarily explore the mechanism of Jiu-long Shi-ba-zheng Decoction in the treatment of gastric ulcer（stomach heat syndrome）,the therapeutic effect of Jiu-long Shi-ba-zheng Decoction on 6-Keto-PGF_1α,TXB₂and IL-8 in serum of rats was evaluated by constructing gastric ulcer（stomach heat syndrome）model in rats.Methods:1.Acute toxicity test:maximum dose method;2.Analgesic test:torsion method and hot plate method;3.Anti-inflammatory test:ear swelling test,capillary permeability test,toe swelling test and cotton ball granuloma test.4.Jiu-long Shi-ba-zheng Decoction for the treatment of gastric ulcer in rats（stomach heat syndrome）model experiment:60 SD rats with body weight of（300±20）g were randomly divided into blank group,model group,positive drug group,Jiu-long Shi-ba-zheng Decoction high-dose,medium-dose and low-dose groups,with 10 rats in each group,male and female in half.Except for the blank group,the other 50 were only used to make the model.The model rats were intragastric with dried ginger decoction twice a day for 14consecutive days.On the 11th day of modeling,15%acetic acid（1m L）was added by intragastric administration once for 4 consecutive days.After successful modeling,medication was given for intervention.The drug administration group was given corresponding drug gavage,while the blank group and model group were given the same amount of distilled water gavage,once a day,for consecutive 7 days.Observation indexes:（1）macroscopic physical signs of rats;（2）Appearance changes of gastric mucosa in each group;（3）Pathomorphological changes of gastric tissues in each group;（4）Ulcer index;（5）Contents of 6-Keto-PGF_1α,TXB₂and IL-8 in serum of rats.Results:1.The maximum tolerance of mice to Jiu-long Shi-ba-zheng Decoction was 270.72g crude drug/kg,which was equivalent to 125.9 times of human dosage.2.Writhing method:Compared with blank group,the number of writhing of mice in each administration group was significantly decreased（P<0.05）,and the effect of medium dose group was the best.Compared with blank group,the incubation period of writhing in positive drug group and medium dose group was significantly prolonged（P<0.05）;Hot plate method:Compared with blank group,pain threshold of mice in each administration group was significantly increased at each period after administration（P<0.05）.3.Ear swelling test:compared with blank group,ear swelling degree of mice in positive drug group,medium and low dose groups was significantly reduced（P<0.05）;Capillary permeability test:compared with blank group,each drug group had obvious inhibitory effect on the exudation of inflammatory substances in mouse abdominal cavity（P<0.05）;Foot swelling test:Compared with model group in the same time phase,the degree of foot swelling in drug groups was significantly decreased（P<0.05）;Cotton ball granuloma test:compared with model group,positive drug group,high and medium dose groups had significant inhibitory effect on granuloma formation（P<0.05）.4.Intervention experiment of Jiu-long Shi-ba-zheng Decoction in gastric ulcer model:the symptoms of gastric heat syndrome were significantly improved,gastric mucosa edema and congestion,inflammatory infiltration of tissue cells were significantly reduced,and ulcer index was significantly decreased（P<0.05）.Compared with the blank group,the serum IL-8 content in the medium dose group had no significant difference,and was significantly increased in the other groups（P<0.01）.Compared with the model group,the serum IL-8 content in the medication group was significantly decreased（P<0.01）.Compared with blank group,the content of 6-Keto-PGF_1αin serum of other groups was significantly decreased（P<0.01）;Compared with model group,all groups except low-dose group were significantly higher than model group（P<0.01 or P<0.05）.Compared with blank group,serum TXB₂in other groups was significantly increased（P<0.01）.Compared with model group,all groups except low-dose group were significantly decreased（P<0.01）.Conclusion（s）:1.There was no acute toxic reaction in mice after intragastric administration of 270.72g crude drug/kg.2.Jiu-long Shi-ba-zheng Decoction has good analgesic and anti-inflammatory effects,and has significant inhibitory effects on both acute and chronic inflammation.3.Jiu-long Shi-ba-zheng Decoction can improve the symptoms of heat syndrome of gastric ulcer,improve gastric ulcer and gastric mucosa edema and congestion,reduce the inflammatory infiltration of tissue cells,and promote the repair of gastric tissue.4.The mechanism of Jiu-long Shi-ba-zheng Decoction for the treatment of gastric ulcer fever syndrome may be that it protects the damaged gastric mucosa by reducing the concentration of damage factors IL-8 and TXA₂,and increases the concentration of protective factor PGI₂.It can reduce vasospasm,platelet aggregation and thrombosis,protect gastric mucosa and promote ulcer healing.
