| Part1Objective: To evaluate the IgE levels of anti-BP180NC16 A antoantibody in sera and blister fluid in bullous pemphigoid patients,and the correlation between them and the severity of the disease.Methods: We evaluate the IgE levels of anti-BP180NC16 A antoantibody in sera in 13 bullous pemphigoid patients and 13 healthy subjects,and in blister fluid in 13 bullous pemphigoid patients and the negative pressure blister fluid in 13 vitiligo patients by ELISA.SPSS25.0 was used for t-test and Pearson correlation coefficient analysis.Results: The positive rate of BP180NC16 A IgE antibody in sera was 69.23%(9/13)and in blister fluid was 84.6%(11/13).BP180NC16 A IgE antibody in sera in BP patients was(312.34±90.39)pg/mL and in healthy subjects was(7.07±0.54)pg/mL by t-test,and the difference was statistically significant(t=3.377,P < 0.05).BP180NC16 A IgE antibody in blister fluid in BP patients was(221.29±68.93)pg/mL and in vitiligo patients was(14.49±3.19)pg/mL by t-test,and the difference was statistically significant((t=2.977,P <0.05).There was no significant difference in BP180NC16 A IgE antibody between sera and blister fluid by paired t-test(t=-0.865,P > 0.05).However,the data showed that in most cases,the antibody in sera was higher than that in blister fluid;Pearson correlation analysis was performed on 13 patients by using SPSS25.0.The correlation coefficient between serum BP180NC16 A IgE antibody and urticaria/erythema score was 0.860,which was significantly correlated at the level of 0.01.There was no correlation between serum BP180NC16 A IgE antibody and BPDAI score and erosion/blister score.There was no correlation between BP180NC16 A IgE antibody and BPDAI score,urticaria/erythema score,and erosion/blister score.The correlation coefficient between BP180NC16 A IgE antibody in serum and in blister fluid was 0.699,a significant correlation at the level of 0.01.Conclusions: For BP patients,a high positive rate of BP180NC16 A IgE antibody was detected by ELISA.The BP180NC16 A IgE antibody in sera and blister fluid in patients was significantly higher than that in control group.There was no significant difference in BP180NC16 A IgE between sera and blister fluid,but in most cases,BP180NC16 A IgE antibody in sera was higher than blister fluid.The level of BP180NC16 A IgE antibody in sera was significantly correlated with urticaria/erythema score.There was a significant correlation between BP180NC16 A IgE antibody in sera and BP180NC16 A IgE antibody in blister fluid.Part2Objective: To evaluate the levels of Eotaxin-1,Eotaxin-2,Eotaxin-3 and RANTES in sera and blister fluid in bullous pemphigoid patients and their correlation with disease activity.Methods: We evaluate the levels of Eotaxin-1,Eotaxin-2,Eotaxin-3 and RANTES in sera in bullous pemphigoid patients and 16 healthy subjects,and in blister fluid in 16 bullous pemphigoid patients and the negative pressure blister fluid in 16 vitiligo patients by ELISA.SPSS25.0 was used for t-test and Pearson correlation coefficient analysis.Results: Eotaxin-1 in sera in BP patients was(701.51±145.48)pg/mL and in healthy subjects was(150.90±28.86)pg/mL by t-test,and the difference was statistically significant(t=3.30,P < 0.05).Eotaxin-1 in blister fluid in BP patients was(497.93±80.65)pg/mL and in vitiligo patients was(9.12±2.30)pg/mL by t-test,and the difference was statistically significant(t=5.35,P < 0.05).Eotaxin-2 in sera in BP patients was(899.1±615.45)pg/mL and in healthy subjects was(108.73±23.37)pg/mL,and the difference was statistically significant(t=3.64,P < 0.05).Eotaxin-2 in blister fluid of BP patients was(1056.36±166.90)pg/mL and in vitiligo patients was(18.95±7.80)pg/mL by t-test,and the difference was statistically significant(t=5.48,P < 0.05).Eotaxin-3 in sera in BP patients were(1346.18±271.44)pg/mL and in healthy subjects was(120.0±12.28)pg/mL,and the difference was statistically significant(t=3.29,P < 0.05).Eotaxin-3 in blister fluid of BP patients was(817.3±130.45)pg/mL and in vitiligo patients was(219.91±30.31)pg/mL by t-test,and the difference was statistically significant(t=4.66,P<0.05).RANTES in sera in BP patients were(35112.5±7610.2)pg/mL and in healthy subjects was(2686.0±1030.5)pg/mL,and the difference was statistically significant(t=3.94,P < 0.05).RANTES in blister fluid of BP patients was(146.5±54.2)pg/mL and in vitiligo patients was(56.8±7.58)pg/mL by t-test,and the difference was not statistically significant(t=1.64,P>0.05).Pearson correlation analysis was performed on 16 BP patients with complete clinical data by SPSS25.0.The correlation coefficient between serum EOS and disease severity was 0.663,which was significantly correlated at the level of 0.01.The correlation coefficient between eotaxin-3 in serum and disease severity was 0.610,which was significant at the level of 0.05.Conclusions: The levels of Eotaxin-1,Eotaxin-2,Eotaxin-3 and RANTES in sera in BP patients were higher than those in control group.Eotaxin-1,Eotaxin-2,and Eotaxin-3 in the blister fluid of BP patients were higher than those in the control group,but there was no significant difference in RANTES between the experimental group and the control group.There was a significant correlation between disease activity and serum EOS and Eotaxin-3 in BP patients,but no significant correlation between Eotaxin-1,Eotaxin-2 and RANTES. |
