Objective Observe the level of IL-17C in the serum of patients with inflammatory bowel diseas(IBD),explore the role of IL-17C in the occurrence and development of the disease,and look for in vitro inflammatory factors suitable for assisting in the diagnosis of IBD.Provide clinical support for seeking the mechanism of IL-17C in IBD patients;Create an animal model,establish a mouse model of inflammatory bowel disease,and preliminarily explore the possible role and regulatory mechanism of IL-17C in the pathogenesis and development of mouse inflammatory bowel disease.Methods 1.Select 32 inpatient/outpatient patients with inflammatory bowel disease in our hospital,collect the fasting clinical serum of the patients;and retrospectively analyze their diagnostic data,including but not limited to age,gender,disease severity and other indicators,and conduct group analysis according to them.Fifty normal healthy subjects were selected as the control subjects,also the subjects were divided into the same age group as the patient group(because our region is dominated by UC patients,this experiment will focus on UC).Routinely collect 3ml of patient serum,use ELISA method to detect the level of IL-17C in serum,use electrochemiluminescence method to detect the level of inflammation expression PCT,and use immunoturbidimetric method to detect the level of serum CRP.2.Animal model construction: Prepare experimental female C57BL/6 mice.Randomly divide them into two groups: one group is the WT normal control group,and the other is the WT DSS experimental group.The DSS model group was given 25 g/L DSS drinking water for 7 days to establish an inflammatory bowel disease model,and the control group was given normal drinking water.During the modeling period,the body weight of the mice was recorded and the body weight changes were observed.After 7 days,the mice were sacrificed,the spleen and the whole colon of the mice were dissected and isolated,the expression and localization of IL-17C in the colon tissue were detected by immunohistochemistry,the level of IL-17C m RNA in the colon tissue of the mice was detected by PCR,and the colon was observed by HE staining the degree of histopathological damage.Results 1.The content of IL-17C in patients with inflammatory bowel disease is higher than that in the healthy group,there is a significant difference(P<0.05),and it increases with age,and there is a significant difference in the age range(P<0.05);there are also differences between PCT and CRP Different degrees of increase,there is a significant difference between the two groups(P<0.05).2.Establish the experimental group of mouse inflammatory bowel disease model.Compared with the normal control group,the results of HE staining showed that the colon tissue of the experimental group was severely damaged and a large number of inflammatory cells infiltrated;the level of IL-17C m RNA in the colon tissue was significantly increased with a significant difference(P<0.05).Conclusion The function of IL-17C is similar to the inflammatory substance CRP,PCT and other effects on IBD patients.It is manifested as increased levels of peripheral blood content and is affected by age.It increases with age.It can be combined with CRP and PCT as a kind of in vitro Auxiliary diagnosis method;IL-17C is highly expressed in mouse inflammatory bowel disease model and is involved in the occurrence and development of IBD disease.
