| ObjectiveThis study included 152 patients with chronic gastritis.Using data mining and network pharmacology methods,the clinical symptoms and syndrome distribution of patients were analyzed,the four qi and five flavors of traditional Chinese medicine were related to meridians,drug association analysis and cluster analysis,and core prescriptions were extracted.The network pharmacology analysis of the core prescriptions was carried out to analyze the main active components,core targets and action pathways of the drugs,so as to provide more basis for the traditional Chinese medicine treatment of chronic gastritis from the aspects of clinical thinking and medication.MethodsAll the cases included in this study were from the outpatient clinic of Du Jianmin’s famous doctor’s studio in Wuhan First Hospital from March2021 to December 2021.Through gastroscopy,they were in line with the diagnosis of chronic gastritis and were treated with oral traditional Chinese medicine,a total of 152 cases..In the first study,the patient’s age,gender,symptoms,tongue and pulse were analyzed in the ancient and modern medical record cloud platform（v2.3.5）by collecting the general information of the case data,clinical symptoms,tongue and pulse,TCM syndromes,Chinese medicine and other information,and organized them into Excel files.,TCM syndromes,frequency distribution of TCM,efficacy of TCM,properties and taste of meridian,and the correlation analysis of TCM,clustering of TCM and complex network analysis of TCM are carried out within the system,and the core prescription for the treatment of chronic gastritis is obtained.Study 2 used the research method of network pharmacology to analyze the mechanism of the core prescription acting on chronic gastritis,searched the active ingredients and protein targets of traditional Chinese medicine through the TCMSP database,and used the Uniprot database to standardize the protein name and find the corresponding gene name.Find chronic gastritis disease targets in disease-related databases such as OMIM,Gene Cards,and DRUGBANK,draw Venn diagrams of drug genes and disease genes,obtain the intersection targets of the two,import String11.0,and perform PPI network analysis of protein interactions.The Cytoscape3.7.1 drawing software draws the network diagram,and analyzes the topological parameters in the software to obtain the key components and targets of the core prescription for the treatment of chronic gastritis.The intersection targets of drugs and diseases were imported into the Metascape database,and enrichment analysis was performed to predict the biological processes and action pathways of core prescriptions in the treatment of chronic gastritis.ResultsStudy one（1）The age and gender distribution of 152 chronic gastritis patients showed that there were 48 male patients,104 female patients,60 patients in the age group of 50-59 years,the largest number,and 40 patients in the age group of 60-69 years old,followed by the number of patients;In the distribution of tongue and pulse,insomnia,dry mouth,bitter mouth,loose stool,abdominal fullness,irritability,acid swallowing,etc.occur more frequently.In terms of tongue texture,there are red tongue,light red tongue,dark red tongue,and light red tongue There are more frequent occurrences of tooth marks,thin white fur,white fur,greasy fur,and less fur on the tongue coating,and more frequent occurrences of stringy,deep and thin pulses in the pulse condition;（3）There are 35 TCM syndromes,There were 32 cases of spleen-stomach deficiency and cold syndrome,and the frequency was the highest;（4）A total of 93 traditional Chinese medicines were used.Keel,oyster,etc.are used in large amounts.Among the four qi and five rank distribution frequencies of traditional Chinese medicine,warm drugs are the most,followed by flat,slightly cold,and mild temperature,and sweet drugs are the most,followed by acrid,bitter,light,and sour.,belong to the spleen meridian and lung meridian the most,and the distribution of the efficacy of traditional Chinese medicine is mainly concentrated in helping yang to transform qi,diuresis and swelling,calming and reducing qi,warming meridian and dredging meridians,invigorating the middle and benefiting qi,sweating and relieving muscle,etc;The main related drug pairs are Guizhi-licorice,tangerine peel-licorice,Guizhi-Atractylodes,Codonopsis-Atractylodes and so on.Cluster analysis found five groups of drugs,the first group: dried ginger,betel nut,the second group: skullcap,chuanxiong,talc,bitter almond,the third group: coix seed,the fourth group: white peony,purple Su Ye,Citrus aurantium,Cyperus officinalis,the fifth group: Guizhi,Astragalus,Codonopsis,Poria,Atractylodes,tangerine peel,licorice.The complex network of drugs analyzed the core prescriptions of tangerine peel,cinnamon twig,astragalus,codonopsis,tuckahoe,white peony,citrus aurantium,Cyperus officinalis,perilla leaves,atractylodes,coix seed and licorice.Study two（1）There are 169 active ingredients of core prescription drugs,of which eicosane-11,14-dienoic acid methyl ester,（+）-catechin,CLR,stigmasterol,hederin,etc.are shared by some drugs There are 265 gene targets of drugs,673 gene targets of diseases,and 106 intersecting targets of drugs and diseases;（2）PPI network analysis of intersecting targets shows IL6,ALB,TNF,AKT1,IL1 B,VEGFA,TP53,JUN,PTGS2,CASP3,MMP9,etc.are important targets,the core prescription-active ingredients-targetsdisease network graph topology analysis,the more important active ingredients are quercetin,isorhamnetin,luteolin,tangerine peel The most important gene targets are PTGS2,NOS2,PPARG,PTGS1,MAPK14,HSP90AA1,F2,NOS3,CHRM3,SLC6A4,etc;（3）Enrichment analysis predicts that the main pathways of core prescriptions in the treatment of chronic gastritis are cancer,PI3K-Akt,IL-17,proteoglycan in cancer,tumor necrosis factor,apoptosis apoptosis,Th17 cell differentiation,Toll-like receptors,HIF-1 and other signaling pathways,etc.The main biological processes include responses to inorganic substances,responses to oxidative stress,responses to bacteria,apoptosis signaling pathways,Lipopolysaccharide reaction,etc.ConclusionThe included 152 patients with chronic gastritis were found through data mining analysis and network pharmacology analysis of clinical data:（1）In this clinical study of chronic gastritis,the clinical manifestations of spleen-stomach deficiency and spleen-stomach-qi deficiency were the main symptoms,with phlegm turbidity,liver For stagnation,most of the medicines use sweet and warm products to help yang transform qi,warm the spleen and stomach,and also relieve dampness and transport spleen,regulate qi and relieve stagnation;（2）The core prescription uses multiple active ingredients,multiple targets,and multiple signaling pathways to treat stagnation.The role of disease is concentrated in many aspects such as inflammation,immunity,cell proliferation and apoptosis,and cancer. |
