Chloranthus fortunei,a perennial herb belonging to the genus Chloranthus in the family Chloranthaceae,is distributed in southern China.C.fortunei been used to alleviate the sufferings of carbuncles,bruises,furuncles,and dysmenorrhea.C.fortunei is an anti-inflammatory drug widely used in Jiangxi Province.It has a long history of folk medicine and is rich in wild medicinal materials.Previous phytochemical investigations on this plant have revealed the presence of sesquiterpenoids,sesquiterpenoid dimers,diterpenoids,flavonoids,coumarins,and phenolic acids.In order to obtain novel sesquiterpenoids,this paper studied the chemical constituents of C.fortunei.The components of C.fortunei were extracted by ethanol immersion and reflux.The chemical constituents of its ethanol extract were separated and purified by silica gel column chromatography,gel chromatography,reversed phase chromatography,high performance liquid chromatography and other chromatography methods.The structure of the substance was identified by modern methods such as mass spectrometry,nuclear magnetic resonance spectroscopy,ultraviolet spectroscopy and so on.Twelve compounds were isolated and purified from its ethanol extract.It is intended to provide substantial evidence for the material basis of the efficacy of C.fortunei.Objective: To develop and utilize the medicinal value of C.fortunei appropriately and study the active components of this traditional Chinese medicine,this research intends to carry out systematic extraction,separation,and structural identification,which will lay a foundation for the pharmacological activity research and comprehensive application of C.fortunei.Methods: A variety of modern natural product separation analysis techniques were used in this project.Firstly,using 95% ethanol solvent to extract the whole herbs of C.fortunei.and obtain the extract,which was then extracted by ether,ethyl acetate in series.Secondly,using all kinds of chromatographic techniques to separate and purify the different extracts,such as silica gel column chromatography,MCI column,Sephadex LH-20,and HPLC.Finally,the structure of isolated monomer compound was identified by modern spectral analysis techniques,such as NMR and MR.Results: Twelve compounds were isolated and Identified from the ethanol extract of C.fortunei,including two undescribed compounds.They were Fortulactone A(1),Fortulactone B(2),Shizukaol F(3),Shizukaol E(4),Shizukaol I(5),Chlorahololide D(6),Chloramultilide B(7),Chlorajaponilide C(8),Chlorajaponilide B(9),Shizukaol P(10),loliolide(11),Istanbulin B(12).Conclusion: This subject further enriches the study on the chemical constituents of Chloranthus.
