Purpose: In this study,a variety of chronic stress stimulation methods were used to construct a chronic unpredictable mild stress(Chronic unpredictable mild stress,CUMS)rat model,and high-intensity interval training on treadmills was used as an intervention method to detect serum BUN,CRP and skeletal muscle IL-15 content and IL-15 mRNA expression.The purpose is to explore(1)the effect of high-intensity interval training on inflammation in depressed rats;(2)analyze the effects of fast and slow muscles on muscle factor IL-15 content and IL-15 mRNA expression in rats under high-intensity interval training.It provides a basis for whether high-intensity interval training can exert an antidepressant effect by affecting the expression of inflammatory factors,and provides a theoretical reference for the formulation of physical exercise training exercise prescriptions.Methods: Forty SD rats at 6 weeks of age were divided into control group(C)and depression group(D),and CUMS was used for four-week modeling.After modeling,they were divided into control group(C),depression group(D),training group(E),and depression + training group(DE),and HIIT was performed for four weeks.Using weekly weight measurement,sugar water preference experiment and open field experiment behavior evaluation method every two weeks to determine the model of depression rats.Immediately after the end of HIIT each week,the serum of the rats in the E group and the DE group was taken,and the serum BUN was detected by a semi-automatic biochemical instrument to determine the training volume.In the ninth week,the rat serum,gastrocnemius(fast muscle)and soleus muscle(slow muscle)were collected 24 hours after the end of the experiment.ELISA was used to detect serum BUN,CRP and skeletal muscle IL-15 content,and RT-PCR was used to detect IL-15 mRNA expression.Results:(1)After four weeks of CUMS stimulation,the weight of rats,the preference rate of sugar and water,the number of vertical standing in the open field experiment,and the staying time of the central grid in group D were significantly lower than those in group C(P<0.01).The residence time of marginal grid in group D was significantly higher than that in group C(P<0.01).(2)After four weeks of HIIT,the body weight of rats in DE group was significantly lower than that in D group(P<0.01),and the sugar water preference rate and the number of vertical standing times of rats in DE group were significantly higher than those in D group(P<0.05,P<0.01).The residence time of the central grid in the DE group has a tendency to increase compared with the D group.The residence time of marginal grid in the DE group was significantly lower than that in the D group(P<0.01).(3)As the HIIT load gradually increases,the BUN value also gradually increases.The weekly BUN value of the DE group was lower than that of the E group.In the last week of HIIT,the BUN value of the DE group was significantly lower than that of the E group(P<0.01).(4)After four weeks of HIIT,compared with group C,the BUN value of group D showed an increasing trend.The BUN value of DE group was significantly lower than that of C group,D group and E group(P<0.01).There was no significant difference in the CRP value between the groups(P>0.05).Compared with group C,the CRP value of group D showed an increasing trend.Compared with group D,the CRP value of DE group had a downward trend.(5)After four weeks of HIIT,compared with the KC(fast muscle)group,the IL-15 content of the KE group and the KDE group showed an increasing trend.Compared with the MC(slow muscle)group,the IL-15 content of the ME group and the MDE group showed a downward trend.Compared with the KC group,IL-15 mRNA in the KE group and the KDE group showed an increasing trend.Compared with the MC group,IL-15 mRNA in the ME group showed a rising trend,and the MDE group increased significantly(P<0.01).The IL-15 mRNA expression of MD group and MDE group was higher than that of MC group and ME group,with very significant difference(P<0.01).After four weeks of HIIT,compared with the KD group,the IL-15 content and IL-15 mRNA expression of the KDE group showed an increasing trend.Compared with the MD group,the IL-15 content and IL-15 mRNA expression of the MDE group showed a downward trend.Conclusions :(1)Various indicators of psychological stress show the success of model building.Mainly manifested as anorexia behavior(weight),anhedonia phenomenon(sugar preference experiment),decreased exploration ability and fear phenomenon(open field experiment).(2)Sports training monitoring shows that the model is successful.The four-week HIIT selected BUN as the training load monitoring indicator,which showed that the HIIT program was feasible and reached the expected experimental design.It was found that the depression + training group showed better training adaptability.(3)HIIT can improve depression symptoms in rats.The main manifestations are alleviation of anhedonia phenomenon(sugar water preference experiment),improvement of exploration ability and reduction of fear phenomenon(open field experiment).(4)Psychological stress exacerbates inflammation to a certain extent,and HIIT can relieve inflammation.The main manifestation is that the BUN and CRP values have a rising trend after CUMS stimulation.The BUN value of the depressed rats after HIIT for four weeks is significantly lower than that of the depression group,and the CRP value has a downward trend.(5)The anti-inflammatory effect of HIIT training is mainly completed by fast muscles.The main manifestation is that compared with the control group,four weeks of HIIT increased the fast muscle IL-15 content and fast muscle IL-15 mRNA expression in the training group and the depression + training group.
