| Objective:This study was investigated the effects of Inonotus Obliquus extract on exercise performance and energy metabolism in male mice,and to discussion of potential mechanism to enhancing sports performance.Methods:Thirty-two male ICR mice were randomly divided into 4 groups,3 IO groups and a Vehicle group:(1)Vehicle group,(2)IO-1X group(824mg/kg/d),(3)IO-2X group(1648mg/kg/d),and(4)IO-3X group(2472mg/kg/d).They were given daily intragastric doses of the corresponding Inonotus Obliquus extract(0.4m L),in which the Vehicle group received the same amount of deionized water.The mice were supplemented with Inonotus Obliquus extract for 40 days,and the experiments were conducted according to the grasping ability of forelimbs,running exhaustion and spontaneous wheel movement.And blood biochemistry,organ index,pathological section,Gas metabolism;The relevant data of the experimental group and the control group were compared and analyzed to comprehensively evaluate the efficacy and dose-dependent effect of the product on exercise performance and energy metabolism.Research Results:(1)Compared with the Vehicle group,The forelimb grip strength of mice in the IO-1X,IO-2X and IO-3X groups was increased by 13.56%,25.34%and 39.31%,respectively,and there was a dose-dependent effect(p=0.0067),and the running time was prolonged by2.39,2.09 and 3.12 times(p=0.0039),respectively.And there is a dose-dependent effect(p<0.0001),the velocity of spontaneous wheel movement increased by 41.26%,49.85%and 32.16%,respectively(p<0.0001),the velocity increase was most significant in the IO-2X group(p<0.0001),indicating that IO supplementation enhanced the muscle strength and endurance of mice,and the exercise ability was enhanced.(2)Compared with the Vehicle group,IO supplementation reduced the levels of blood Ammonia(AMM)and creatine kinase(CK)(p=0.0021,p<0.0001),increased the levels of glucose(GLU)and free fatty acid(FFA)(p<0.0001,p=0.0068),and the mean dose-dependent increase of liver glycogen and muscle glycogen water(p<0.0001,p=0.00259),played an anti-fatigue role.In addition,the level of total cholesterol(TC)in IO group decreased(p<0.0001),the level of triglyceride(TG)decreased(p=0.0002),and all had dose-dependent effects,indicating that IO could reduce blood lipid and regulate lipid metabolism.(3)Compared with the Vehicle group,the weight of epididymis fat pad in IO group was significantly decreased(p=0.0226),and the weight of hind limb gastrocnemius and relative brown fat weight were significantly increased(p=0.0053,p=0.0011),among which the weight of hind limb gastrocnemius was significantly increased in IO-2X group(p=0.0054).After body composition,free fat mass(FFM)increased significantly(p<0.0001),with the most significant weight gain in the IO-2X group(p=0.0054,p<0.0001),and IO supplementation had a dose-dependent effect on epididymal fat pad,relative brown fat weight,and FFM in mice(p=0.0053,p=0.0021,p=0.0021).(4)The results of metabolism:IO supplementation significantly increased the average oxygen uptake(Avg_VO2)and the average energy expenditure(Avg_EE)in mice.Compared with the Vehicle group,the difference of average respiratory quotient(Avg_RQ)in IO-2X group was the most significant,and there was a dose-dependent effect(p<0.0001p<0.0001,p<0.0001),indicating that IO supplementation can regulate resting metabolism,improve energy metabolism,and improve exercise endurance.(5)Histomorphological examination.No pathological features were found in sections between each group.Therefore,in this study,IO supplementation for 40 days was safe for mice.Research Conclusions:(1)Inonotus Obliquus extract can alleviate fatigue,protect muscle from injury and increase energy reserve by reducing blood ammonia level,reducing creatine kinase accumulation,increasing blood glucose and free fatty acid content and glycogen reserve,and improving body composition,thus improving exercise performance of mice.(2)Inonotus Obliquus extract had positive regulation effect on lipid metabolism by improving blood lipid level,reducing respiratory quotient and increasing brown fat content in mice.Increased muscle mass and oxygen uptake indicate increased energy expenditure,improved energy utilization,and regulation of resting metabolism.Therefore,Inonotus Obliquus extract improved energy metabolism in mice.Therefore,Inonotus Obliquus extract may be a potential natural supplement to relieve fatigue,improve exercise performance.IO-2X(1648 mg/kg BW/d)can be used as a reference dose to provide scientific clues for athletes to improve exercise performance. |
