| Purpose: We investigate the effects of salidroside supplementation on exercise performance and biochemical index in eccentric trained mice.Methods: Forty-eight 6-week-old male ICR mice were randomly divided into 6groups: sedentary control group(SC),eccentric exercise training group(ECC),1-fold salidroside group(SA1X,410mg/kg/day),2-fold salidroside group(SA2X,820mg/kg/day),1-fold salidroside combined with eccentric exercise training group(SA1X+ECC),2-fold salidroside combined with eccentric exercise training group(SA2X+ ECC).Mice were treated for 4 weeks with salidroside supplementation and eccentric training with a running platform slope of-20% and a speed of 17 m/min for 30 min per day,five days per week.After 4 weeks of experimentation,a 15 min swimming experiment was performed to detect fatigue indicators in mice: Lactate dehydrogenase(LDH),Ammonia(Amm),glucose(Glu),nonesterified fatty acid(NEFA),and creatine kinase(CK).Forelimb grip strength was measured in mice.The treadmill exhaustion experiment was performed with a 10% incline,gradually increasing the time and speed until exhaustion,and the time of exhaustion was recorded.At the end of the experiment,mice were measured for body composition,muscle glycogen and liver glycogen content,and biochemical values including: alanine aminotransferase(ALT),aspartate aminotransferase(AST),lactate dehydrogenase(LDH),alkaline phosphatase(ALP),plasma albumin(ALB),creatine kinase(CK),creatinine(CRE),total protein(TP),triglycerides(TG),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),and blood glucose(GLU).Results:1.There was no interaction effect between salidroside supplementation and eccentric exercise training on LDH,CK activity,blood ammonia,blood glucose,and free fatty acid content in mice after 15-minute swimming training(p>0.05).Salidroside supplementation had a significant effect on reducing CK activity(p<0.01),and eccentric exercise training had a significant effect on reducing LDH activity and free fatty acid content(p<0.01,p<0.05).The CK value of SA1 X and SA2 X was significantly lower than that of the SC group(p<0.05,p<0.01),and the CK value of SA1X+ECC and SA2X+ECC group was significantly lower than that of the SC group(p<0.01,p<0.05).The LDH activity of ECC and SA2X+ECC groups was significantly lower than that of the SC group(p<0.05,p<0.01).2.Supplementation of salidroside and eccentric exercise training had no significant interaction effect on prolonging the time to exhaustion in mice(p>0.05),but had a significant interaction effect on enhancing forelimb grip strength in mice(p<0.05).Salidroside had a significant effect on prolonging the time to exhaustion in mice(p<0.05).Eccentric exercise training had a significant effect on prolonging the time to exhaustion in mice(p<0.0001).The time to exhaustion in SA1 X,ECC,SA1X+ECC,SA2X+ECC group was significantly higher than that in the SC group(p<0.0001,p<0.01,p<0.01,p<0.01),the effect of increasing the exhaustive exercise time of mice in the SA1 X group is better than that in the SA2 X group.The forelimb grip strength of SA1X+ECC and SA2X+ECC groups was significantly higher than that of the SC group(p<0.05,p<0.0001),and the grip strength of SA2X+ECC group was significantly higher than that of SA1X+ECC group(p<0.05).3.Supplementation with salidroside and eccentric exercise training had a significant interactive effect on reducing the relative weight of forelimb muscle,epididymal fat,and brown adipose tissue(p<0.01,p<0.01,p<0.05).The relative weight of forelimb muscles in SA1 X,SA2X and ECC groups was lower than that in the SC group(p<0.05,p<0.01,p<0.05).The relative weight of epididymal fat in SA1 X,SA2X,ECC,SA1X+ECC,SA2X+ECC group was lower than that in the SC group(p<0.0001,p<0.01,p<0.0001,p<0.0001,p<0.0001).4.Supplementation with salidroside and eccentric exercise training had a significant interactive effect(p<0.01)on increasing relative lean body mass.Salidroside has a significant effect on reducing relative fat content(p<0.01),and eccentric exercise training has a significant effect on reducing relative fat content(p<0.05).The relative lean body mass of SA1 X,SA2X,ECC,SA1X+ECC,SA2X+ECC group was significantly higher than that of SC group(p<0.01,p<0.01,p<0.0001,p<0.01,p<0.0001),and the relative fat mass was higher than that of SC group.Significantly lower in the SC group(p<0.01,p<0.05,p<0.01,p<0.01,p<0.01).5.Supplementation with salidroside and eccentric exercise training had a significant interactive effect on increasing muscle glycogen content(p<0.0001).Salidroside had a significant effect on increasing liver glycogen content(p<0.0001).Eccentric exercise training had a significant effect on increasing liver glycogen content(p<0.05).The liver glycogen content of SA2 X,SA1X+ECC,SA2X+ECC group was significantly higher than that of SC group(p<0.01,p<0.01,p<0.0001).SA2 X group had better effect on increasing liver glycogen content of mice than SA1 X group,The effect of salidroside supplementation combined with eccentric exercise training in increasing liver glycogen in mice was better than that of eccentric exercise training alone.The muscle glycogen content of SA1 X,SA2X,ECC,SA1X+ECC,SA2X+ECC group was significantly higher than that of the SC group(p<0.01,p<0.0001,p<0.01,p<0.05,p<0.05),and the SA2 X group had muscle glycogen content was significantly higher than SA1 X group(p<0.0001).6.Supplementation of salidroside and eccentric exercise training had a significant interactive effect on reducing the levels of AST,TG,TC,and LDL-C(p<0.01,p<0.0001,p<0.01,p<0.0001).Salidroside had a significant effect on increasing TP content(p<0.0001).The AST levels of SA1 X,SA2X,ECC,SA1X+ECC group were significantly lower than the SC group(p<0.01,p<0.01,p<0.01,p<0.05).TG(p<0.0001,p<0.0001,p<0.0001,p<0.0001,p<0.01),TC(p<0.01,p<0.01,p<0.01)0.0001,p<0.05,p<0.01),and LDL-C(p<0.01,p<0.01,p<0.0001,p<0.01,p<0.01)levels of SA1 X,SA2X,ECC,SA1X+ECC,SA2X+ECC group were significantly lower than those in SC group.Conclusion: The combination of salidroside supplementation and eccentric exercise training can relieve exercise fatigue in mice,prolong the running exhaustion time of mice,enhance the grasping force of the forelimbs of mice,increase the content of muscle glycogen and liver glycogen,and reduce the relative fat content of mice,increased the relative lean body mass of mice,and decreased the levels of AST,TG,TC,and LDL-C in mice.And the combination of salidroside supplementation and eccentric exercise training in improving the exercise performance of mice was better than that of eccentric exercise training alone. |
