Penicillium cicada is a kind of parasitic fungus of Cordyceps sobolifera,while Cordyceps sobolifera is a traditional Chinese medicine(TCM),which has the function of antitumor activity and immunomodulatory.It has been used for diabetes,kidney disease and so on.Paecilomyces cicadae has attracted considerable attention because the content of active ingredients is no less than that of natural cicada.At present,research for artificial cultivation,submerged fermentation instead of natural cicada has become a research direction to many researchers.However,there's almost no research about compatibility of Penicillium cicada with traditional Chinese medicine for research.Chinese traditonal medicines were used as medicinal substance to replace the traditional fungal culture(bagasse,wheat bran,and other agricultural products),the potency substance in the fermentation process provides nutrients such as carbonitride which fungal requires,the enzyme that produced by the fungus can change the component of herbs,achieve better efficacy,and reduce the toxicity of raw herbs.Radix Astragali is one of the commonly used TCM,which has significant immunomodulatory and antitumor activity,it is used to treat liver cirrhosis,diabetes and heart disease clinically.It's active ingredients include the composition such as flavonoids,polysaccharides and astragalus saponin,which make Radix Astragali good for health care,thus,its output and dosage are huge.Penicillium Cicada and Radix Astragali both have the effect of immune regulation,their medicinal properties are nearly the same.In this research,Penicillium cicada-Radix Astragali bi-directional fermentation system is established.The main active ingredient content such as polysaccharide,flavonoids and total saponins were determined,what's more,we conducted pharmacodynamic test to assess it's medical properties,so as to provide a new way for environmental protection and efficient utilization of Radix Astragali residue.The main research contents of this study are as follows:(1)Use single-factor experiment to evaluate the optimal fermentation technology(fermentation temperature,inoculation quantity,fermentation time)of submerged fermentation through comparison of biomass in order to lay foundation for Penicillium cicada/astragalus bi-directional fermentation system.(2)Established the Paecilomyces cicadae-Radix Astragali and Paecilomyces cicadaeRadix Astragali residue bi-directional fermentation system,optimized condition of bidirectional fermentation system orthogonal design method with index of cordyceps acid content.Measure the content total sugar,polysaccharide,flavonoids and total saponins of different fermentation time by chemical colorimetric method.The results shows that:With the fermentation goes on,Paecilomyces cicadae translated the nutrients of Radix Astragali and it's residue,which confirmed that the fermentation system is feasible and worth studying.(3)According to the change of active in gredients,we conducted the anti-oxidation,immune enhancement and antitumor efficacy research to explore the change of efficacy.The results shows that:DPPH free radical clearance rate and the ferrous ion chelating ability are improved after fermentation both in Paecilomyces cicadae-Radix Astragali and Paecilomyces cicadae-Radix Astragali residue bi-directional fermentation system.Both of the two kinds of bi-directional fermentation system can promote the neutral red phagocytosis of macrophage,The inhibition of coarse polysaccharide extract of fermented Radix Astragali and residue on MCF-7 cell were improved compared with unfermented sanmples,moreover,the result illustrates that there's a concentration-dependent between the inhibition percentage of MCF-7 cell and polysaccharide extract.The results show that it's meaningful to use fungus to deal with Radix Astragali and it's residue.After fermentated by Penicillium Cicada,active ingredient content of Radix Astragali changed significantly,and it's pharmacological activities have been significantly enhanced,which shows that fermented Radix Astragali have better medicinal value.As to the residue of Radix Astragali,although it's content of active ingredient is lower than fermentated Radix Astragali,its active ingredient improved significantly and has a good pharmacological activities after fermentation.Hence,Radix Astragali residue has a good application prospect in the field of medicine and veterinary medicine after bi-directional fermentation.
