Trace elements are vital to the health of humans and animals.Under healthy conditions,the trace elements that make up the organism maintain a balance with each other to maintain normal life functions.Trace element imbalance,electrolyte disorder and heavy metal poisoning are important reasons for destroying the steady-state balance of the animal body.In recent years,the growth-promoting and health-care effects of trace elements on animals have gradually attracted attention.There are many types of trace elements added in feed,and the phenomenon of excessive supply of trace elements and nutritional imbalance in modern animal husbandry production is widespread.At the same time,there are significant antagonistic,synergistic and interaction effects among trace elements in animals.Combining the above factors,a variety of factors that lead to trace element imbalance,electrolyte disorder and heavy metal poisoning still exist,and still threaten animal health.Therefore,it is necessary to establish an animal homeostasis imbalance model in order to systematically and accurately identify the effects of trace elements and electrolyte homeostasis on the animal body.In this experiment,10 healthy female small-tailed Han sheep at the age of 12 months were selected from a ranch in Heilongjiang,and the 10 ewes were divided into two groups:a control group and an EDTA group using the method of self-control.In order to establish repeated blood collection and long-term infusion venous access,and improve the efficiency of continuous dynamic blood collection and intravenous administration,the modified Sedinger technique was used to insert the central venous catheter into the sheep's external jugular vein before the start of the experiment.Before the start of the experiment,whole blood of sheep was collected as the control group before treatment;the experimental group was intravenously injected with 4%concentration of metal ion chelator EDTA(pH 7.4,60 mL/h)480 mL,injected daily for 8 hours,and continuously injected intravenously for 3 days to construct sheep Trace element and electrolyte imbalance model.An electrolyte and blood gas analyzer was used to determine K+,Na+,Cl-,Ca2+,pH and other related indicators in sheep blood.ICP-MS was used to detect 53kinds of elements including Ca,Cu,Na,Mg,P,K and Zn in sheep serum.The results of the study showed that the central venous catheterization of sheep is convenient for real-time collection of sheep blood,and it is less irritating to sheep during blood collection.After 3 days of continuous intravenous injection of 4%metal ion chelating agent EDTA,the expression levels of ALB,AST and TB in sheep serum were significantly increased(P<0.05);the contents of An Gap,Na+,K+and Cl-in sheep blood were significantly lower than The control group(P<0.01);the content of Li,Be,Na,Br,Y,Ru and Re in the sheep serum of the EDTA group was significantly lower than that in the control group(P<0.05),P,K,Zn,As,Rb,In and The content of Cs was extremely significantly lower than that of the control group(P<0.01);the content of Ca,Sr and Pd in the serum of sheep in the EDTA group was significantly higher than that of the control group(P<0.05),and the content of Cu was extremely significantly higher than that of the control group(P<0.01)).The above results suggest that the concentration of some mineral elements and electrolyte ions in the blood of sheep has changed significantly after intravenous injection of EDTA.Intravenous injection of EDTA can induce the steady-state imbalance of mineral elements in sheep,in order to find out the effects of trace elements and electrolyte homeostasis on the animal body Provide a reliable animal model.
